目的:研究白藜芦醇( Resveratrol,Res)对妊娠期糖尿病大鼠血糖、血脂和相关炎症因子的影响。方法取60只妊娠5 d SD大鼠,通过一次性腹腔注射链脲佐菌素( STZ)35 mg/kg制备妊娠期糖尿病大鼠模型,随机分为模型组、白藜芦醇60 mg/kg组、白藜芦醇120 mg/kg组、白藜芦醇240 mg/kg组和盐酸二甲双胍(200 mg/kg)组,每组12只;另取12只妊娠5 d SD大鼠作为正常妊娠组,12只非妊娠雌性大鼠作为对照组。分别于给药前和给药第7,14天检测空腹血糖和胰岛素水平。给药满2周后,测量各组大鼠体质量;通过全自动生化检测仪测定总胆固醇( TC)、三酰甘油( TG)、低密度脂蛋白( LDL-C)、高密度脂蛋白( HDL-C)及C反应蛋白( CRP)水平,采用ELISA法测定血浆中肿瘤坏死因子-α( TNF-α)和白细胞介素-6(IL-6)水平。结果正常妊娠组和对照组空腹血糖和胰岛素水平、体质量、TC、TG、LDL-C、HDL-C 、CRP、TNF -α和IL-6水平比较差异均无统计学意义( P均>0.05)。与正常妊娠组相比,模型组胰岛素水平显著降低,空腹血糖水平显著升高,体质量明显增加,血清TC、TG、LDL-C水平显著升高,血清HDL-C水平显著降低,血浆CRP、TNF-α、IL-6水平显著升高,差异均有统计学意义(P均<0.05)。与模型组相比,白藜芦醇120 mg/kg组、白藜芦醇240 mg/kg组胰岛素水平显著升高,血糖水平,体质量,血清TC、TG、LDL-C水平及血浆CRP、TNF -α、IL-6水平均显著降低,差异均有统计学意义(P均<0.05);白藜芦醇240 mg/kg组血清HDL-C水平显著升高(P<0.05)。与盐酸二甲双胍组相比,白藜芦醇240 mg/kg组胰岛素水平显著升高,血清TC、TG、LDL-C水平及血浆CRP、TNF-α、IL-6水平均显著降低,差异均有统计学意义(P均<0.05)。结论白藜芦醇对妊娠期糖尿病大鼠具有剂量依赖性的提高胰岛素分泌、降低血糖和体质量、调节血脂和炎症因子水平的作用,并且改善胰岛素分泌及调节血脂和炎症因子水平的作用强于盐酸二甲双胍。

목적:연구백려호순( Resveratrol,Res)대임신기당뇨병대서혈당、혈지화상관염증인자적영향。방법취60지임신5 d SD대서,통과일차성복강주사련뇨좌균소( STZ)35 mg/kg제비임신기당뇨병대서모형,수궤분위모형조、백려호순60 mg/kg조、백려호순120 mg/kg조、백려호순240 mg/kg조화염산이갑쌍고(200 mg/kg)조,매조12지;령취12지임신5 d SD대서작위정상임신조,12지비임신자성대서작위대조조。분별우급약전화급약제7,14천검측공복혈당화이도소수평。급약만2주후,측량각조대서체질량;통과전자동생화검측의측정총담고순( TC)、삼선감유( TG)、저밀도지단백( LDL-C)、고밀도지단백( HDL-C)급C반응단백( CRP)수평,채용ELISA법측정혈장중종류배사인자-α( TNF-α)화백세포개소-6(IL-6)수평。결과정상임신조화대조조공복혈당화이도소수평、체질량、TC、TG、LDL-C、HDL-C 、CRP、TNF -α화IL-6수평비교차이균무통계학의의( P균>0.05)。여정상임신조상비,모형조이도소수평현저강저,공복혈당수평현저승고,체질량명현증가,혈청TC、TG、LDL-C수평현저승고,혈청HDL-C수평현저강저,혈장CRP、TNF-α、IL-6수평현저승고,차이균유통계학의의(P균<0.05)。여모형조상비,백려호순120 mg/kg조、백려호순240 mg/kg조이도소수평현저승고,혈당수평,체질량,혈청TC、TG、LDL-C수평급혈장CRP、TNF -α、IL-6수평균현저강저,차이균유통계학의의(P균<0.05);백려호순240 mg/kg조혈청HDL-C수평현저승고(P<0.05)。여염산이갑쌍고조상비,백려호순240 mg/kg조이도소수평현저승고,혈청TC、TG、LDL-C수평급혈장CRP、TNF-α、IL-6수평균현저강저,차이균유통계학의의(P균<0.05)。결론백려호순대임신기당뇨병대서구유제량의뢰성적제고이도소분비、강저혈당화체질량、조절혈지화염증인자수평적작용,병차개선이도소분비급조절혈지화염증인자수평적작용강우염산이갑쌍고。
ABSTRACT:Objective It is to investigate the effects of Resveratrol(Res) on the levels of blood glucose and lipid and correlated inflammatory cytokines in Gestational Diabetic rats.Methods 60 rats with gestation for five days were injected STZ (35 mg/kg) by intraperitonal to make gestational diabetic model, which were randomly devided into five groups:normal gesta-tion group, gestational diabetic model group, Resveratrol(60, 120 and 240 mg/kg) treated groups and Metformin hydrochlo-ride 200 mg/kg treated group, every group had 12 rats.Anther 12 rats with gestation for five days as normal gestation control group and 12 rats with no gestation as normal control group.Before the drugs were given and 7, 14 days after treated, the level of plasma glucose and insulin were determined.After two weeks’ treatment, the body weight were detected, the content of TC, TG, LDL-C, HDL-C, CRP in serum were detected with automated analysis, and the level of TNF-α, IL-6 in plas-ma were also detected with ELISA.Results Compared with normal control group, the level of plasma glucose and insulin of the normal gestation control group was not significantly different(P>0.05), the body weigh were not significantly different(P>0.05), the content of TC, TG, LDL-C, HDL-C and the level of CRP, TNF-α, IL-6 were also not significantly different (P>0.05).Compared with the normal gestation control group, the level of plasma glucose in gestational diabetic model group was significantly increased and the level of insulin was significantly decreased(P<0.01); the body weight was significantly increased ( P<0.01);the content of TC, TG, LDL-C were significantly increased and the HDL-C was significantly de-creased(P<0.05, P<0.01).Compared with model group, the content of TC, TG, LDL-C, the level of plasma glucose, CRP,TNF-α,IL-6 body weight were significantly decreased(P<0.05, P<0.01), while the level of plasma insulin was decreased in Res (120, 240 mg/kg) treated groups, furthermore, the content of HDL-C in Res 240 mg/kg treated group was significantly increased(P<0.05).Compared with the Metformin hydrochloride treated group, the content of TC, TG, LDL-C and the levels of CRP, TNF-α, IL-6 in plasma were significantly decreased while plasma insulin was increased in Res (240 mg/kg) treated group.Conclusion Res had dose-dependent effect of improving insulin secretion, lowering blood sugar and body weight, regulating plasma lipid, the level of CRP、TNF-α、IL-6 and these effects of Res was better than Metformin hydrochloride except lowering blood sugar and body weight.
