目的 分析miR-153在卵巢癌中的表达,并探讨miR-153的生物学作用.方法 荧光定量 PCR 法检测卵巢癌组织及细胞中 miR-153 的表达水平;转染 miR-153 类似物(mimics)至SKOV3细胞,MTT法、平板克隆实验及流式细胞仪分别检测miR-153对细胞增殖及凋亡的影响.结果miR-153在卵巢癌组织及细胞中的表达显著下调;转染miR-153 mimics后细胞生长受到明显抑制,克隆数目减少,细胞早期凋亡率显著增加(P<0.01).结论 miR-153在卵巢癌中表达下调;过表达miR-153可显著抑制SKOV3细胞增殖,促进细胞凋亡.

목적 분석miR-153재란소암중적표체,병탐토miR-153적생물학작용.방법 형광정량 PCR 법검측란소암조직급세포중 miR-153 적표체수평;전염 miR-153 유사물(mimics)지SKOV3세포,MTT법、평판극륭실험급류식세포의분별검측miR-153대세포증식급조망적영향.결과miR-153재란소암조직급세포중적표체현저하조;전염miR-153 mimics후세포생장수도명현억제,극륭수목감소,세포조기조망솔현저증가(P<0.01).결론 miR-153재란소암중표체하조;과표체miR-153가현저억제SKOV3세포증식,촉진세포조망.
Objective This study aims to analyze the miR-153 expression profile in ovarian cancer, explore the biological functions of miR-153.Methods The relative expression of miR-153 in ovarian cancer tissues and cancer cell lines were detected by real-time quantitative RT-PCR. Introduction of miR-153 mimics to SKOV3 cells. The cell proliferation ability were detected by MTT assay and colony formation, and cell apoptosis was detected by flow cytometry.Results The expression of miR-153 was significantly decreased in ovarian cancer tissues and cell lines; introduction of miR-153 mimics into the SKOV3 cells inhibited cell proliferation, colony formation ability and induced apoptosis (P<0.01). Conclusion miR-153, which is significantly downregulated in ovarian cancer tissues and cell lines, inhibits cell proliferation and induces apoptosis.
