目的:观察重度子痫前期患者胎盘组织Notch1、Jagged1 mRNA和蛋白表达变化,并探讨其意义。方法选择重度早发型子痫前期患者为A组,重度晚发型子痫前期患者为B组,正常孕妇为C组,各25例。均行剖宫产,取胎盘组织,用实时荧光定量PCR法检测胎盘组织Notch1、Jagged1 mRNA,用免疫组化法检测胎盘组织Notch1、Jag-ged1蛋白。结果与C组相比, A、B组Notch1、Jagged1 mRNA表达水平降低( P均<0.05);与B 组相比, A组Notch1、Jagged1 mRNA表达水平降低(P均<0.05)。 Notch1、Jagged1蛋白主要连续表达于绒毛外滋养细胞胞质及血管内皮细胞胞质。 A、B、C组Notch1蛋白阳性表达率分别为36.67%、50.00%、83.33%,Jagged1蛋白阳性表达率分别为43.33%、50.00%、83.33%,Notch1、Jagged1蛋白阳性表达率A组低于B组,B组低于C组,P均<0.05。结论 Notch1、Jagged1在重度子痫前期患者胎盘组织中呈低表达,二者可能参与重度子痫前期尤其是重度早发型子痫前期的发生。

목적:관찰중도자간전기환자태반조직Notch1、Jagged1 mRNA화단백표체변화,병탐토기의의。방법선택중도조발형자간전기환자위A조,중도만발형자간전기환자위B조,정상잉부위C조,각25례。균행부궁산,취태반조직,용실시형광정량PCR법검측태반조직Notch1、Jagged1 mRNA,용면역조화법검측태반조직Notch1、Jag-ged1단백。결과여C조상비, A、B조Notch1、Jagged1 mRNA표체수평강저( P균<0.05);여B 조상비, A조Notch1、Jagged1 mRNA표체수평강저(P균<0.05)。 Notch1、Jagged1단백주요련속표체우융모외자양세포포질급혈관내피세포포질。 A、B、C조Notch1단백양성표체솔분별위36.67%、50.00%、83.33%,Jagged1단백양성표체솔분별위43.33%、50.00%、83.33%,Notch1、Jagged1단백양성표체솔A조저우B조,B조저우C조,P균<0.05。결론 Notch1、Jagged1재중도자간전기환자태반조직중정저표체,이자가능삼여중도자간전기우기시중도조발형자간전기적발생。
Objective To observe the expression changes of Notch 1 and Jagged1 mRNA and protein in the placentas from patients with severe preeclampsia and to investigate the significance .Methods Twenty-five patients with early-onset severe preeclampsia were recruited as the group A , and 25 patients with the late-onset severe preeclampsia were recruited as the group B .Meanwhile , 25 healthy pregnant women who delivered after 38 weeks were defined as group C .The real-time fluorescent quantitative PCR was used to investigate the expression of Notch 1 and Jagged1 protein and mRNA in the placentas .Results Compared with group C , the expression levels of Notch 1 mRNA and Jagged1 mRNA were decreased in the group B and group A (all P<0.05).The expression levels of Jagged1 mRNA and Notch1 mRNA in the group A were decreased as compared with those of the group B (all P<0.05).Notch1 and Jagged1 proteins were expressed in cytoplasm of villous trophoblasts continuously and vascular endothelial cells .The positive expression rates of Notch 1 protein were 36.67%, 50.00%and 83.33%in the group A, group B and group C.The positive rates of Jagged1 and Notch1 in the group A were lower than those in the group B, and that in the group B was lower than that in the group C (all P<0.05). Conclusion The Notch1 and Jagged1 was lowly expressed in patients with severe preeclampsia , which indicated that the down-regulated expression may take part in the occurrence of the early-onset severe preeclampsia .
