目的:观察应用兔脂肪干细胞(ADSCs)诱导的成骨细胞复合组织工程骨支架材料---猪小肠黏膜下层(SIS)在体外构建组织工程骨膜的生物学特性。方法从兔脂肪中分离ADSCs,在条件培养基下诱导分化为成骨细胞,然后将成骨细胞与SIS进行体外复合培养,检测细胞的生物学特性,并对细胞复合而成的生物活性骨膜进行碱性磷酸酶(ALP)检测。观察ADSCs与SIS复合后的生长增殖和诱导分化的情况。结果脂肪组织可获得ADSCs,贴壁后各组细胞均呈短梭形。贴壁细胞CD44阳性表达,CD29阴性表达。细胞生长曲线结果显示,细胞与SIS复合后均具有活>的增殖能力。ALP检测显示,确定复合至组织工程骨膜上的细胞可以成骨。结论 ADSCs诱导的成骨细胞与SIS复合构建组织工程骨膜具有良好的成骨特性。

목적:관찰응용토지방간세포(ADSCs)유도적성골세포복합조직공정골지가재료---저소장점막하층(SIS)재체외구건조직공정골막적생물학특성。방법종토지방중분리ADSCs,재조건배양기하유도분화위성골세포,연후장성골세포여SIS진행체외복합배양,검측세포적생물학특성,병대세포복합이성적생물활성골막진행감성린산매(ALP)검측。관찰ADSCs여SIS복합후적생장증식화유도분화적정황。결과지방조직가획득ADSCs,첩벽후각조세포균정단사형。첩벽세포CD44양성표체,CD29음성표체。세포생장곡선결과현시,세포여SIS복합후균구유활>적증식능력。ALP검측현시,학정복합지조직공정골막상적세포가이성골。결론 ADSCs유도적성골세포여SIS복합구건조직공정골막구유량호적성골특성。
Objective To observe the biological characteristics of tissue engineered periosteum constructed by adipose-derived stem cells (ADSCs) from rabbit combined with bone tissue engineering scaffold - small intestinal submucosa (SIS) in vitro. Methods The adipose-derived stem cells were separated from rabbit adipose and induced osteoblast ad-hered on conditioned medium, osteoblast and SIS were proliferated, the active cellular function was detected. The cells composition generated biological periosteum was detected alkaline phosphatase (ALP). The grow tendency and induction differentiation of ADSCs and combined with SIS were observed. Results ADSCs were gotten from rabbit adipose, and ADSCs after adherence showed short fusiform in vitro. The cells after adherence showed CD44 positive expression and CD29 negative expression. The results of growth curve showed osteoblast induced from ADSCs on SIS had active multi-plica tion capacity. The results of ALP detection showed osteoblast induced from ADSCs on SIS had similar osteogenic potential. Conclusion The tissue-engineered periosteum constructed by osteoblast induced from ADSCs on SIS showed good osteogenesis characteristics.
