目的:观察自拟柴郁寄生汤对高催乳素血症( HPRL)模型大鼠卵巢组织PRL/PRLR-JAK2-STAT5信号传导通路的影响及其改善卵巢功能的机制。方法 Wistar大鼠60只皮下注射多巴胺受体阻断剂灭吐灵造成高催乳素血症模型。造模成功后按拉丁方法随机分为模型组、溴隐亭组、大剂量组、中剂量组、小剂量组各10只,并设正常组10只。中药高剂量组柴郁寄生汤剂量为18 g· kg-1· d-1,中剂量组为9 g· kg-1· d-1,低剂量组为4#.5 g· kg-1· d-1。各药物组每只大鼠每次灌胃量均为2 ml。溴隐亭组给溴隐亭2 mg · kg-1· d-1;模型组及正常组依据体质量予等容量蒸馏水灌胃;各组每日给药1次,连续给药30 d。治疗结束后将大鼠处死,量取大鼠卵巢体积;检测血清雌二醇( E2)、促黄体生成素( FSH);通过Western blot及PCR法检测卵巢STAT、PRLR表达水平,以观察高催乳素血症环境对PRL/PRLR-JAK2/STAT5信号传导通路及卵巢功能的影响,并探讨其可能机制。结果与正常组比较,模型组大鼠血清PRL水平明显升高, E2、FSH水平均明显下降( P <0.05);卵巢STAT、PRLR蛋白表达水平均明显降低( P <0.05)。与模型组比较,各治疗组血清PRL水平均明显下降,血清E2、FSH水平及卵巢STAT、PRLR表达水平均明显升高。不同治疗组间比较,中药中剂量组大鼠血清E2、FSH水平改善最显明,卵巢STAT、PRLR蛋白表达明显最强( P <0.05),而PRL水平差异均无统计学意义( P >0.05)。结论柴郁寄生汤治疗高催乳素血症对改善卵巢功能有良好疗效,其作用机制可能是调节卵巢PRL/PRLR-JAK2/STAT5信号传导通路相关因子的表达,进而改善E2、FSH等生殖激素水平。

목적:관찰자의시욱기생탕대고최유소혈증( HPRL)모형대서란소조직PRL/PRLR-JAK2-STAT5신호전도통로적영향급기개선란소공능적궤제。방법 Wistar대서60지피하주사다파알수체조단제멸토령조성고최유소혈증모형。조모성공후안랍정방법수궤분위모형조、추은정조、대제량조、중제량조、소제량조각10지,병설정상조10지。중약고제량조시욱기생탕제량위18 g· kg-1· d-1,중제량조위9 g· kg-1· d-1,저제량조위4#.5 g· kg-1· d-1。각약물조매지대서매차관위량균위2 ml。추은정조급추은정2 mg · kg-1· d-1;모형조급정상조의거체질량여등용량증류수관위;각조매일급약1차,련속급약30 d。치료결속후장대서처사,량취대서란소체적;검측혈청자이순( E2)、촉황체생성소( FSH);통과Western blot급PCR법검측란소STAT、PRLR표체수평,이관찰고최유소혈증배경대PRL/PRLR-JAK2/STAT5신호전도통로급란소공능적영향,병탐토기가능궤제。결과여정상조비교,모형조대서혈청PRL수평명현승고, E2、FSH수평균명현하강( P <0.05);란소STAT、PRLR단백표체수평균명현강저( P <0.05)。여모형조비교,각치료조혈청PRL수평균명현하강,혈청E2、FSH수평급란소STAT、PRLR표체수평균명현승고。불동치료조간비교,중약중제량조대서혈청E2、FSH수평개선최현명,란소STAT、PRLR단백표체명현최강( P <0.05),이PRL수평차이균무통계학의의( P >0.05)。결론시욱기생탕치료고최유소혈증대개선란소공능유량호료효,기작용궤제가능시조절란소PRL/PRLR-JAK2/STAT5신호전도통로상관인자적표체,진이개선E2、FSH등생식격소수평。
Objective To observe the effect of Chaiyujisheng decoction on hyperprolactinemia ( hPRL) model rat o-varian tissue PRL/PRLR-JAK2-STAT5 signaling pathway and improvement of the ovarian function mechanism.Methods Wistar rats were injected with dopamine receptor blockers to cause a high level of prolactin (60 rats).The model group was randomly divided into model group, bromocriptine group, large dose group, middle dose group, small dose group, and the normal group with 10 rats in each group.Traditional Chinese medicine high dose group’ s Chaiyujisheng decoction’ s amount was 18 g· kg-1 · d-1 ,medium dose group was 9 g· kg-1 · d-1 , and low dose group was 4.5 g· kg-1 · d-1 .Each time, the amount of each rat were fed with 2 ml.Bromocriptine group were given bromocriptine 2 mg· kg-1 · d-1;model group and normal group according to body mass were given equal volume of distilled water by gavage.Each group administered once dai-ly, continuous administration of 30 d.At the end of treatment, rats were sacrificed, rat ovarian volume were measured;detec-ted serum estradiol ( E2 ) , luteinizing generated hormone ( FSH) , through the method of Western blot and PCR to detect ovari-an’ s STAT and PRLR expression level to observe the effects of hyperprolactinemia environment on PRL/PRLR-JAK2-STAT5 signaling pathway and ovarian function, and to explore its possible mechanism.Results Compared with the normal group, model group rats’ serum PRL level increased significantly, E2 , FSH levels were significantly decreased ( P <0.05);ovari-an’s STAT and PRLR protein expression level were significantly decreased ( P <0.05).Compared with model group,in the treatment group, the serum PRL levels were significantly decreased and serum E2 , FSH levels and ovarian’ s STAT and PRLR expression levels were significantly increased.Between the different treatment groups, middle dose of traditional Chinese medi-cine group rats’ serum E2 and FSH levels improvement were the most obviously, and ovarian’ s STAT and PRLR protein ex-pression was the highest(P<0 ?.05), and differences in the level of PRL had no statistical significance ( P >0.05).Conclu-sion It demonstrated that the Chaiyujisheng decoction could improve ovarian function in hyperprolactinemia, its mechanism maybe due to regulate the expression of ovarian PRL/PRLR-JAK2-STAT5 signaling pathway related factors, so as to improve E2 , FSH’ s level.
