目的:观察用人包皮成纤维细胞( hPFF)支持生长的人胚胎干细胞( hESC )的体内全能性。方法将第20代hPFF支持生长的hESC和hPFF分别接种到2只严重联合免疫缺陷的小鼠(SCID)小鼠体内,分别为观察组和对照组,观察5周内两组小鼠一般状态及是否有畸胎瘤形成。取观察组小鼠畸胎瘤制作组织学切片,验证其是否包含了源于全部3个胚层的各种分化细胞。结果观察组SCID小鼠接种后第5天即可在接种部位触到包块,包块进行性增大。观察组小鼠包块理组织学检查显示,该肿块包含3个胚层的组织结构,具备畸胎瘤的病理组织学特点。结论用hPFF支持生长的hESC有体内分化全能性。

목적:관찰용인포피성섬유세포( hPFF)지지생장적인배태간세포( hESC )적체내전능성。방법장제20대hPFF지지생장적hESC화hPFF분별접충도2지엄중연합면역결함적소서(SCID)소서체내,분별위관찰조화대조조,관찰5주내량조소서일반상태급시부유기태류형성。취관찰조소서기태류제작조직학절편,험증기시부포함료원우전부3개배층적각충분화세포。결과관찰조SCID소서접충후제5천즉가재접충부위촉도포괴,포괴진행성증대。관찰조소서포괴리조직학검사현시,해종괴포함3개배층적조직결구,구비기태류적병리조직학특점。결론용hPFF지지생장적hESC유체내분화전능성。
Objective To observe the totipotency analysis of human embryonic stem cells ( hESCs) supported by hu-man postnatal foreskin fibroblasts (hPFFs).Methods The hESCs of the 20th passage supported by hPFF and hPFF were respectively injected into the hind legs of the severe combined immunode? cient mice ( SCID) and taken as the observation and control group .After 5-week, we observe the general state and teratoma formation of the two mice .The teratoma was taken from the observation group for the histological analysis to investigate whether there were different kinds of differentia -ted cells derived from three germ layers .Results We touched the masses in the vaccination site of SCID mice on the fifth day after inoculation , and the masses increased progressively .Five weeks later , we killed the mice to strip the tumor , and then they received the histologic examination .It showed that the mass contained three germ layers'structures with the histo-logic characteristics of teratoma .Conclusions The hESCs supported by hPFFs have totipotency .
