试验以杜洛克猪、大白猪、大围子猪、沙子岭猪、桃源黑猪、五指山猪和杜长大商品猪为试验材料,通过测序和PCR-RFLP的方法对胰岛素诱导基因2(INSIG2)5'调控区域的多态性进行检测,并分析该基因对猪肉质性状的影响,结果表明:在该区域发现T-502C、G-460C和T-609C 3个突变位点,T-502C和G-460C所选所有品种都处于Hardy-Weinberg平衡状态,T-609C除了五指山猪外也都符合Hardy-Weinberg平衡,在肉质性状上,T-502C位点,TT型个体的系水力和嫩度显著高于CC型;G-460C位点,杂合子CG型在贮存损失上显著高于CC型和GG型,大理石纹GG型和CG型差异不显著但是都显著高于CC型,肉质色度方面GG型和CC型差异不显著但显著高于CG型;T-609C位点,TT型的贮存损失显著高于CT型, 但和CC型差异不显著, 嫩度CC型显著高于CT型和TT型,INSIG2基因可能是一个与猪肉系水力、嫩度和大理石纹相关的分子遗传标记.

In this experiment, the Duroc, Yorkshire, Daweizi, Shaziling, Taoyuan, Wuzhishan and the DLY Hybrid were chose, the polymorphism of insulin induced genes 2 (INSIG2)'s 5' regulatory regionswere detected through the sequencing and PCR-RFLP method, and we also analyzed the association of polymorphisms of INSIG2 with the meat traits, the results show that this area has three mutations:T-502C, G-460C and T-609C, the T-502C and G-460C arein Hardy Weinberg equilibrium among all the selected varieties, T-609C are also in Hardy Weinberg equilibrium besides wuzhishan pigs; on the meat quality traits, for T-502C sites, water holding capacity and tenderness of TT type are significantly higher than that of CC type; for G-460C sites, CG type is significantly higher than CC and GG, in marble, GG and CG type was not significant but were significantly higher than that of CC, on fleshy chromaticity, GG type and CC was not significant, but significantly higher than CG type; for T-609C sites, TT is significantly higher than CT in storage loss, but no significant difference between CC type, in tenderness, CC was significantly higher than that of CT and TT, INSIG2 gene is likely to be one of the molecular genetic markers forwater holding capacity, tenderness and the marble.
