柚皮苷是中药骨碎补的主要活性成分,有促进成骨和骨组织新陈代谢的功能,目前载柚皮苷的羟基磷灰石/壳聚糖人工骨缓释材料未见报道.制备了壳聚糖(chitosan,CS)质量分数为10%,20%和30%的壳聚糖/羟基磷灰石-柚皮苷(chitosan/hydroxyapa-tite-naringin,CS/HA-N)缓释材料,并以 SEM、IR 和XRD 对材料进行了表征,同时研究了材料的机械性能、体外降解行为以及药物缓释行为.IR 和 XRD 结果表明,缓释材料的无机相为部分碳酸化的低结晶度羟基磷灰石,与天然骨相似.SEM 结果显示材料中羟基磷灰石(hydroxyapatite-N,HA)呈片状结晶.机械性能测试结果表明,缓释材料具有良好的力学性能, CS 含量为10%,20%和30%的 CS/HA-N 抗压强度分别为17.45±3.42,16.86±3.7和19.34±4.69 MPa.随着缓释材料中 CS 含量的增大,材料吸水率随之增大.体外缓释及体外降解行为研究结果表明,CS/HA对柚皮苷有良好的缓释效果,其药物释放速率以及降解速率随壳聚糖含量的增大而增大.CS/HA-N 作为载药人工骨替代材料具有良好的应用前景.

Naringin was the main active ingredient in rhizoma drynariae,a traditional Chinese medicine herb, which has the function of promoting osteogenesis and bone metabolism.So far,Naringin loaded hydroxyapatite/chitosan composite for bone analogues has not been reported.In this paper,the chitosan/hydroxyapatite-naring-in (CS/HA-N)composite with 10%,20% and 30% chitosan contain were prepared.The samples were charac-terized by SEM,IR and XRD.Their mechanical properties,in-vitro release behavior and in-vitro degradation behavior were studied as well.The results showed that the crystals of this CS/HA-N composite containing car-bonate has low crystallinity,which was similar to natural bone.SEM results showed that the hydroxyapatite (hydroxyapatite-N,HA)presented plate shaped crystals.Mechanical performance test results showed that the material had good mechanical properties,the compressive strength of 10%,20%,30% CS/HA-N materials were 17.45±3.42,1 6.86±3.73,1 9.34±4.69 MPa,respectively.With chitosan content increasing in composite, water absorption rate of CS/HA-N was getting higher.The results of in-vitro release and in-vitro degradation test showed that CS/HA-N had good sustained release effect.The drug release rate and the degradation rate in-creased with the increase of chitosan content in CS/HA-N.CS/HA-N had good application prospect as drug de-livery systems for bone tissue engineering.
