目的:从 IL-4/ STAT6信号通路入手,以该通路负反馈调节因子 SOCS1和关键因子 STAT6为检测指标,多角度探查溃疡性结肠炎的发生与 IL-4的关系。方法将实验动物分为6组,采用免疫法造模,用免疫组化法检测大鼠 SOCS1、STAT6的活性。结果模型组实验大鼠 SOCS1活性(0.4855±0.1780)显著低于正常组(0.6552±0.1874),P <0.05;而 STAT6活性(0.8030±0.2082)则明显高于正常组(0.5893±0.1919), P <0.05。四逆散能显著增加实验大鼠 SOCS1活性(0.5765±0.2080)(P <0.05),降低 STAT6活性(0.5798±0.1526)(P <0.05);柴芍配伍(0.5625±0.2757)对 SOCS1活性的影响与四逆散(0.5765±0.2080)比较差异无统计学意义,对 STAT6活性(0.5294±0.1443)表达的影响显著优于四逆散(0.5798±0.1526)(P <0.05);芍枳甘配伍(0.6169±0.2006)对 STAT6活性的影响作用明显,与四逆散(0.5798±0.1526)比较差异无统计学意义;柴枳甘配伍对 SOCS1(0.5091±0.2279)、STAT6(0.7561±0.2636)的影响与模型组(0.4855±0.1780;0.8030±0.2082)比较差异无统计学意义。结论四逆散能够通过刺激 SOCS1活性,抑制 STAT6活性表达来调节 IL-4/ STAT6通路,进而干预实验性溃疡性结肠炎。其中,柴芍配伍作用明显;芍枳甘配伍对 STAT6活性的影响作用明显,但对 SOCS1的影响不显著;柴枳甘配伍对 SOCS1、STAT6的影响均不显著。

Objective To explore the relation with IL-4 and UC through multi angles,starting with the IL-4 / STAT6 pathway. Methods The experimental animals were divided into 6 groups. We made the model in immune method,de-tected activity of SOCS1 and STAT6 in rats with immune. Results The activity SOCS1 in model group of rats(0. 485 5 ± 0. 178 0)were significantly lower than that of normal group(0. 655 2 ± 0. 187 4)(P < 0. 05),but the activity of STAT6(0. 803 0 ± 0. 208 2)were significantly higher than that in the normal group(0. 589 3 ± 0. 191 9) (P < 0. 05). Sinisan Powders can significantly increase the activity of SOCS1(0. 576 5 ±0. 208 0)in experimental rats (P <0. 05),decrease STAT6 activity(0.579 8 ±0.152 6)(P <0. 05). The effects of Chaishao compatibility(0. 562 5 ± 0.275 7)was no significant difference with SOCS1 and Sinisan(0. 576 5 ±0. 208 0)Powders(P >0. 05),the activity of STAT6(0.529 4 ±0.144 3)was significantly better than the Sinisan(0.579 8 ±0.152 6)Powders(P <0.05). Shaozhigan compatibility effects on activity of STAT6(0. 616 9 ± 0. 200 6)significantly. Chaizhigan compatibility had no significant difference with SOCS1(0.509 1 ±0.227 9),STAT6(0.756 1 ±0.263 6)and the model group(0.485 5 ±0.178 0;0.803 0 ±0.208 2)in effects(P >0.05). Conclusion Sinisan Powders can regulate expression of the IL-4/ STAT6 pathway by stimulating the activity of SOCS1,inhibiting the activity of STAT6,then intervention experiment UC. And the effect of Chaishao compatibility was the best obvious;the effects of Shaozhigan compatibility was obvious on activity of STAT6,but not on SOCS1. The effect of Chaizhigan compatibility were not obvious on SOCS1 and STAT6.
