从甘肃定西某淀粉加工厂附近土壤中分离得到一株产普鲁兰酶酶源菌 AI-1,通过形态学、生理生化试验及16S rRNA 序列鉴定并对其进行系统发育分析,鉴定为解淀粉芽孢杆菌(Bacillus amyloliquefaciens),对其发酵培养基成分和发酵条件进行了优化。优化后的发酵培养基成分为:可溶性淀粉1.5%,酵母膏1%,蛋白胨1%,NaCl 0.5%,K2 HPO40.1%,MgSO4·7H2 O 0.05%;最佳发酵条件为:培养温度36℃,发酵培养基初始 pH 7.0,接种量8%(V/ V),摇床转速150 r/ min,发酵周期72 h。在此优化条件下,菌株 AI-1发酵所产普鲁兰酶的酶活由最初的2.45 U/ mL提高到了4.52 U/ mL。

A strain AI-1 having Pullulanase activity was isolated from the soil near a starch-processing fac-tory in Dingxi,Gansu province. The optimal culture conditions for enzyme production were determined. Based on 16S rRNA sequence homology analysis and phylogenetic analysis. AI-1 was identified as Bacillus amyloliquefaciens,with its physiological and biochemistry characters. And then its fermentation medium composition and fermentation conditions were optimized,the optimum fermentation medium composition were as follows:soluble starch 1. 5% ,yeast extract 1% ,peptone 1% ,NaCl 0. 5% ,K2 HPO4 0. 1% ,Mg-SO4 ·7H2 O 0. 05% . Under the optimum fermentation condition of initial 36 ℃,pH 7. 0,150 r/ min and an inoculum size of 8%(V/ V)for 72 h,the activity of Pullulanase produced by AI-1 reached the peak at 4. 52 U/ mL from the initial activity of 2. 45 U/ mL.
