目的:探讨γ-干扰素(IFN-γ)对哮喘小鼠气道炎症和肺组织中MUC5ac 表达的影响。方法:40只雌性C57bL/6小鼠随机分为正常组、哮喘组、IFN-γ5万IU组和IFN-γ10万IU组,每组10只。测定各组小鼠呼吸道阻力,刘氏染色法分类计数支气管肺泡灌洗液(BALF)中细胞数,ELISA 法检测 BALF 中 IL-13的浓度,Ab-PAS 染色计数气道上皮杯状细胞,PCR 检测肺组织中MUC5ac mRNA 表达。结果:与正常组相比较,哮喘组小鼠呼吸道阻力、BALF 中细胞数,IL-13浓度明显增加,肺组织中 MUC5ac 表达升高(P <0.05);IFN-γ能减轻呼吸道阻力、BALF中细胞数,降低BALF中IL-13浓度和肺组织中MUC5ac 表达(P <0.05)。结论: IFN-γ能调节哮喘小鼠Th1/Th2平衡,抑制杯状细胞增生和肺组织中MUC5ac mRNA 的表达。

목적:탐토γ-간우소(IFN-γ)대효천소서기도염증화폐조직중MUC5ac 표체적영향。방법:40지자성C57bL/6소서수궤분위정상조、효천조、IFN-γ5만IU조화IFN-γ10만IU조,매조10지。측정각조소서호흡도조력,류씨염색법분류계수지기관폐포관세액(BALF)중세포수,ELISA 법검측 BALF 중 IL-13적농도,Ab-PAS 염색계수기도상피배상세포,PCR 검측폐조직중MUC5ac mRNA 표체。결과:여정상조상비교,효천조소서호흡도조력、BALF 중세포수,IL-13농도명현증가,폐조직중 MUC5ac 표체승고(P <0.05);IFN-γ능감경호흡도조력、BALF중세포수,강저BALF중IL-13농도화폐조직중MUC5ac 표체(P <0.05)。결론: IFN-γ능조절효천소서Th1/Th2평형,억제배상세포증생화폐조직중MUC5ac mRNA 적표체。
Objective To investigate the effects of IFN-γon airway inflammation and protein MUC5ac mRNA expression in mice with asthma. Methods Forty c57bL/6 mice were randomly divided into control group, model group, IFN-γ 50,000IU group and IFN-γ 100,000IU group, 10 for each group. The change rate of airway resistance was detected after acetylcholine chloride provocation. The count of inflammatory cells in BALF was analyzed. Levels of IL-13 in BALF were measured by ELISA. The lung tissue sections were stained with Ab-PAS for identification of goblet cells proliferation. Expressions of MUC5ac mRNA in the lung tissues were determined by real time-PCR. Results As compared with the control group , the change rate of airway resistance , count of inflammatory cells , levels of IL-13 and MUC5ac mRNA in lung were significantly increased in the model group (P < 0.05). IFN-γ reduced airway resistance, inflammatory cells, levels of IL-13 and MUC5ac mRNA in the lungs (P < 0.05). Conclusions IFN-γ can regulate the balance of Th1/Th2, inhibit airway goblet cell proliferation , and protein MUC5ac mRNA expression in mice withasthma.
