目的 用米非司酮构建黄体功能不全( luteal phase defect , LPD)大鼠模型,通过比较性腺轴各部位的形态学变化,讨论补肾助孕方对LPD大鼠下丘脑-垂体-卵巢-子宫性腺轴形态结构的改善作用. 方法 将大鼠分为空白组、模型组以及中药低、高剂量组共4组,每组10只,空白组给予等渗盐水1 mL/100 g,模型组给予米非司酮1 mL/100 g,中药低、高剂量组分别予中药0 .4及1 .2 mL/100 g. 观察下丘脑-垂体-卵巢-子宫各部位的形态结构变化. 结果 ①电镜下模型组下丘脑可见线粒体嵴断裂,空泡化,粗面内质网脱颗粒,核膜溶解,分泌颗粒量少;②光镜下模型组垂体细胞分布密度稍稀,部分细胞核固缩,细胞质出现空泡化;电镜下模型组大鼠垂体前叶细胞中细胞核明显皱缩变形,核染色质凝聚,细胞器排列紊乱,线粒体内腔肿胀,扩张形成空泡状;③光镜下模型组卵巢中闭锁卵泡增多,颗粒细胞层减少,黄体数量少,中药干预后黄体数量增多, 体积增大,卵泡颗粒细胞层增厚;④光镜下模型组子宫内膜层变薄, 管壁子宫腺体数量减少,腺腔小,间质胶原纤维增多. 中药干预后子宫内膜层增厚,子宫腺体数量增加,腺腔扩大. 结论 用米非司酮构建LPD大鼠模型,其下丘脑-垂体-卵巢-子宫轴上的下丘脑、垂体、子宫、卵巢的形态结构均发生了改变,补肾助孕方可对上述改变起到改善的作用.
目的 用米非司酮構建黃體功能不全( luteal phase defect , LPD)大鼠模型,通過比較性腺軸各部位的形態學變化,討論補腎助孕方對LPD大鼠下丘腦-垂體-卵巢-子宮性腺軸形態結構的改善作用. 方法 將大鼠分為空白組、模型組以及中藥低、高劑量組共4組,每組10隻,空白組給予等滲鹽水1 mL/100 g,模型組給予米非司酮1 mL/100 g,中藥低、高劑量組分彆予中藥0 .4及1 .2 mL/100 g. 觀察下丘腦-垂體-卵巢-子宮各部位的形態結構變化. 結果 ①電鏡下模型組下丘腦可見線粒體嵴斷裂,空泡化,粗麵內質網脫顆粒,覈膜溶解,分泌顆粒量少;②光鏡下模型組垂體細胞分佈密度稍稀,部分細胞覈固縮,細胞質齣現空泡化;電鏡下模型組大鼠垂體前葉細胞中細胞覈明顯皺縮變形,覈染色質凝聚,細胞器排列紊亂,線粒體內腔腫脹,擴張形成空泡狀;③光鏡下模型組卵巢中閉鎖卵泡增多,顆粒細胞層減少,黃體數量少,中藥榦預後黃體數量增多, 體積增大,卵泡顆粒細胞層增厚;④光鏡下模型組子宮內膜層變薄, 管壁子宮腺體數量減少,腺腔小,間質膠原纖維增多. 中藥榦預後子宮內膜層增厚,子宮腺體數量增加,腺腔擴大. 結論 用米非司酮構建LPD大鼠模型,其下丘腦-垂體-卵巢-子宮軸上的下丘腦、垂體、子宮、卵巢的形態結構均髮生瞭改變,補腎助孕方可對上述改變起到改善的作用.
목적 용미비사동구건황체공능불전( luteal phase defect , LPD)대서모형,통과비교성선축각부위적형태학변화,토론보신조잉방대LPD대서하구뇌-수체-란소-자궁성선축형태결구적개선작용. 방법 장대서분위공백조、모형조이급중약저、고제량조공4조,매조10지,공백조급여등삼염수1 mL/100 g,모형조급여미비사동1 mL/100 g,중약저、고제량조분별여중약0 .4급1 .2 mL/100 g. 관찰하구뇌-수체-란소-자궁각부위적형태결구변화. 결과 ①전경하모형조하구뇌가견선립체척단렬,공포화,조면내질망탈과립,핵막용해,분비과립량소;②광경하모형조수체세포분포밀도초희,부분세포핵고축,세포질출현공포화;전경하모형조대서수체전협세포중세포핵명현추축변형,핵염색질응취,세포기배렬문란,선립체내강종창,확장형성공포상;③광경하모형조란소중폐쇄란포증다,과립세포층감소,황체수량소,중약간예후황체수량증다, 체적증대,란포과립세포층증후;④광경하모형조자궁내막층변박, 관벽자궁선체수량감소,선강소,간질효원섬유증다. 중약간예후자궁내막층증후,자궁선체수량증가,선강확대. 결론 용미비사동구건LPD대서모형,기하구뇌-수체-란소-자궁축상적하구뇌、수체、자궁、란소적형태결구균발생료개변,보신조잉방가대상술개변기도개선적작용.
Objective The purpose of this study was to construct a rat model of luteal phase defect ( LPD) with mifepristone ( RU486 ) and investigate the improving effect of Bushen Zhuyun Recipe ( BZR ) on the gonad axis morphology of the LPD rat . Meth ods We established rat models of LPD by giving RU486 at 1 mL/100 g followed by treatment with BZR at 0.4 (medium dose) or 1.2 mL/100g ( high dose ) .Then we observed the changes of morphology of the hypothalamus pituitary ovary uterus . Results Electron microscopy revealed hypothalamic mitochondria vacuolization , degranulation of rough endoplasmic reticulum , dissolved nuclear membrane , and decreased secretory granules , while light microscopy exhibited slightly reduced density of pituitary cells , karyopyknosis of some of the cells ,and cytoplasmic vacuolization in the model rats .Electron microscopy also showed obvious shrinkage of cell nuclei , chromatin condensation , disordered cell arrangement , swelling mitochondrial cavity , and formation of vacuoles in the pituitary anterior lobe, while light microscopy also manifested increased ovarian follicular atresia , reduced granule cell layers and corpora lutea , thinned endometrial layers , decreased uterine glands of the tube wall , reduced glandular cavity , and increased interstitial collagen fibers .After BZR treatment, the number and volume of corpora lutea were increased , the granulosa cell layer and endometrial layer thickened , the number of uterine glands increased , and the gland cavity expanded . Conclusion The axes of the hypothalamus , pituitary, uterus,and o -varian were morphologically changed in the RU 486-induced rat models of LPD, while BZR could improve these changes .
