Xinshubao%myocardial infarction%apop tosis
目的:探讨心舒宝对大鼠心肌梗死的保护作用及其药理机制,为临床用药提供理论依据。方法:0.25 g/mL 心舒宝灌胃二周后诱导大鼠心肌梗死模型,二周后进行大鼠心肌组织的 Masson’s Trichrome 染色实验;利用胰酶消化方法分离大鼠心脏原代内皮细胞,并培养于培养液。用0.005%,0.01%,0.05%的心舒宝处理细胞后,缺氧培养24 h、48 h,流式细胞仪分析其凋亡率。结果:心舒宝处理组的大鼠心肌梗死面积及其切面胶原明显小于对照组(P <0.01);0.005%心舒宝处理的大鼠心脏原代内皮细胞在缺氧48 h 后,其凋亡明显降低(P <0.01)。结论:心舒宝可能通过降低大鼠心脏原代内皮细胞的凋亡而发挥保护大鼠心肌梗死的作用。

목적:탐토심서보대대서심기경사적보호작용급기약리궤제,위림상용약제공이론의거。방법:0.25 g/mL 심서보관위이주후유도대서심기경사모형,이주후진행대서심기조직적 Masson’s Trichrome 염색실험;이용이매소화방법분리대서심장원대내피세포,병배양우배양액。용0.005%,0.01%,0.05%적심서보처리세포후,결양배양24 h、48 h,류식세포의분석기조망솔。결과:심서보처리조적대서심기경사면적급기절면효원명현소우대조조(P <0.01);0.005%심서보처리적대서심장원대내피세포재결양48 h 후,기조망명현강저(P <0.01)。결론:심서보가능통과강저대서심장원대내피세포적조망이발휘보호대서심기경사적작용。
Objective:To investigate the the protective effect and Pharmacological mechanism of Xinshubao on myocar-dial infarction of rats ,and provide a theoretical basis for clinical use.Methods:The model of myocardial infarction rats were induced by 0.25 g/mL Xinshubao after two weeks,then cardiac muscle Masson’s Trichrome staining was done u-sing trypsin digest and to isolate the rat heart primary endothelial cells,and cultured the cells in 1 640 medium.Flow cytometry analysis the apoptosis rate after Hypoxia culture 24 h,48 h of cells treated with 0.005%,0.01 %,0.05%Xinshubao for 24,28 h.Results:Xinshubao treated rat myocardial infarction area and the section of collagen was signifi-cantly less than the control group (P <0.01 );0.005% of Xinshubao treated rat heart primary endothelial cells in hy-poxic for 48 h,apoptosis Octreased significantly (P <0.01 ).Conclusion:Xinshubao may protect myocardial infarction rats by inhibiting apoptosis of rat heart primary endothelial cells.
