目的:研究原儿茶酸对大鼠心肌缺血/再灌注(myocardial ischemia/reperfusion ,M I/R)损伤的保护作用。方法采用结扎SD大鼠冠状动脉建立M I/R (30 min/3 h)损伤模型,将动物随机分为假手术组、模型组、原儿茶酸组(低、中、高剂量)、丹参多酚酸盐6组。除模型组于再灌注时给予生理盐水外,原儿茶酸低、中、高剂量组及丹参多酚酸盐组于再灌注即刻给予原儿茶酸(5,10和20 mg · kg-1)及丹参多酚酸盐(20 mg · kg -1)。实验过程中监测心功能,实验末测定心肌梗死面积,并检测血清中肌酸激酶同工酶(CK‐MB)、肌钙蛋白(cTnI)水平。结果与模型组相比,原儿茶酸中、高剂量组大鼠心功能得以改善(P<0.05),心肌梗死面积显著减少(P<0.05),血清中心肌损伤酶CK‐MB、cTnI的水平显著下降(P<0.05)。结论原儿茶酸对大鼠MI/R损伤具有一定的保护作用。

목적:연구원인다산대대서심기결혈/재관주(myocardial ischemia/reperfusion ,M I/R)손상적보호작용。방법채용결찰SD대서관상동맥건립M I/R (30 min/3 h)손상모형,장동물수궤분위가수술조、모형조、원인다산조(저、중、고제량)、단삼다분산염6조。제모형조우재관주시급여생리염수외,원인다산저、중、고제량조급단삼다분산염조우재관주즉각급여원인다산(5,10화20 mg · kg-1)급단삼다분산염(20 mg · kg -1)。실험과정중감측심공능,실험말측정심기경사면적,병검측혈청중기산격매동공매(CK‐MB)、기개단백(cTnI)수평。결과여모형조상비,원인다산중、고제량조대서심공능득이개선(P<0.05),심기경사면적현저감소(P<0.05),혈청중심기손상매CK‐MB、cTnI적수평현저하강(P<0.05)。결론원인다산대대서MI/R손상구유일정적보호작용。
Objective To investigate the protection effect of protocatechuic acid on myocardial ischemia/reperfusion (MI/R) injury in rats .Methods The rats were subjected to MI/R (30 min/3 h) by occluding the left anterior descending artery .The rats random‐ly received one of the following treatments intravenously and were randomly divided into six groups :sham group ,model group , protocatechuic acid group (low ,middle and high doses) and salvianolate group .Sham‐operated control rats (sham) underwent the same surgical procedures with the exception of left anterior descending coronary artery occlusion .Protocatechuic acid and salviano‐late groups were treated with the dose of 5 ,10 and 20 mg · kg -1 of protocatechuic acid and 20 mg · kg -1 of salvianolate ,respec‐tively .The change of cardiac function was monitored by multi‐channel physiology recorder .The area of myocardial infarction and the levels of CK‐MB ,cTnI in serum were determined to evaluate the protection effect of protocatechuic acid .Results Compared with the model group ,the cardiac function of MI/R rats was improved(P<0 .05) ,the infarct area and the levels of creatine kinase‐MB (CK‐MB) and cardiac troponin (cTnI) in the middle and high dose of protocatechuic acid groups were significantly decreased (P<0 .05) .Conclusion Protocatechuic acid has excellent cardioprotective effects against MI/R injury in rats .
