Taxus wallichiana Zucc .%Taxus meadia%taxol%cephalomannine
目的:测定并比较维吾尔药材喜马拉雅红豆杉与人工栽培品种曼地亚红豆杉中紫杉醇和三尖杉宁碱的含量。方法采用RP‐HPLC色谱法梯度洗脱,色谱柱为 Century SIL C18‐EPS 柱(250 mm ×4.6 mm ,5μm );流动相为乙腈‐水;流速为1.0 mL · min-1;检测波长为227 nm ;柱温为30℃。结果紫杉醇回归方程:Y=2.050975×107 X+5531.887,线性范围为0.042~1.69μg(r=0.9999),平均回收率为98.0%,RSD=0.98%(n=6);三尖杉宁碱回归方程Y=1.806535×107 X+3046.378,线性范围为0.040~1.6μg(r=0.9999),平均回收率为96.6%,RSD=2.97%(n=6),喜马拉雅红豆杉叶中三尖杉宁碱平均含量高于茎中含量,曼地亚红豆杉相反;两者叶中紫杉醇含量高于茎中含量,而曼地亚红豆杉茎叶中紫杉醇含量并不低于喜马拉雅红豆杉。结论该方法简便、准确、适用于红豆杉药材的质量评价。

Objective To determine and compare the contents of taxol and cephalomannine between Taxus wallichiana Zucc .and Taxus meadia by RP‐HPLC .Methods The isolation was performed on a Century SIL C18‐EPS column (250 mm × 4 .6 mm ,5μm) ,with the mobile phase consisting of acetonitrile‐water ,the flow rate of 1 .0 mL · min-1 ,detection wavelength of 227 nm . The column temperature was 30 ℃ .Results The regression equation of taxol was Y=2 .050 975 × 107 X+5 531 .887 ,linear range within 0 .042‐1 .69 μg(r=0 .999 9) .The average recovery was 98 .0% ,RSD =0 .98% (n=6);the regression equation of cephalo‐mannine was Y=1 .806 535 × 107 X+3 046 .378 ,linear range within 0 .040‐1 .6μg(r=0 .999 9) .The average recovery was 96 .6% ,RSD=2 .97% (n=6) .The cephalomannine content in leaves of Taxus wallichiana Zucc was higher than the average content of stem ,Taxus meadia;The taxol content in leaves was higher than the content in stem ,and the taxol content in Taxus meadia leaf was not lower than that in Taxus wallichiana Zucc .Conclusion The chromatography method is simple ,accurate ,and it is suitable for the quality evaluation of Taxus .
