目的 探讨低氧环境下红景天与角质细胞生长因子/缺氧诱导因子( KGF/HIF)修饰的骨髓间充质干细胞( MSCs)联合应用对大鼠脊髓损伤( SCI)修复的影响. 方法 低氧条件下制备大鼠SCI模型,模型制备成功随机分为4组,对照组给予生理盐水,红景天组给予红景天胶囊,移植组移植KGF/HIF修饰的MSCs,联合应用组同时给予红景天及干细胞移植,评价4组大鼠运动能力恢复情况,观察损伤组织病理学及脊髓神经细胞变化. 结果 移植组、红景天组和联合应用组大鼠联合行为学评分均高于对照组( P<0. 05 );脊髓损伤组织细胞数目增多,神经细胞坏死减少,HIF表达水平升高,尤以联合应用组变化明显. 结论 红景天联合KGF/HIF修饰的骨髓MSCs可促进SCI修复.

Objective To investigate the effect of Rhodiola Rosea combined with kinetics gain factor (KGF) and hypoxia-inducible factor 1 (HIF-1) mediated bone marrow mesenchymal stem cells (MSCs) under hypoxia condition in repair of spinal cord injuries ( SCI) in rats. Methods The SCI rat models were established under hypoxia condition and were randomly divided into 4 groups after successful establishment. The control group was treated with physiological saline;the Rhodiola Rosea group was treated with Rhodiola Rosea capsules; the transplantation group was transplanted with rats' bone marrow MSCs modulated by KGF and HIF;the combination group was transplanted with rats' bone marrow MSCs modulated by KGF, HIF and Rhodiola Rosea. The recovery of motor ability of models was evaluated in each group, and the changes of histopathology and spinal nerve cells were observed in each group. Results In transplantation, Rhod-iola Rosea and combination groups, the combined ethology scores were higher (P<0. 05), SCI histiocytes numbers were larger, numbers of neural apoptosis were decreased, and HIF expression levels were increased higher than those of control group;the changes were more obvious in combination group. Conclusion Rhodiola rosea combined with KGF and HIF may promote the SCI repair modulated by bone marrow MSCs.
