目的 研究胆汁酸对高脂诱导肥胖小鼠血糖和体重的影响,并分析其安全性. 方法 32 只雄性C57BL/6小鼠,随机分为高脂饮食组(HFD组)、高脂饮食加0. 1% w/w胆酸干预组(0. 1 CA组)、高脂饮食加0. 25%w/w胆酸干预组(0. 25 CA组)和高脂饮食加0. 25%熊去氧胆酸干预组(UDCA组),每组8只. 干预后测定血糖、胰岛素、总胆汁酸、丙氨酸转氨酶( ALT)和天冬氨酸转氨酶( AST)的水平,采集肝脏组织称重并进行组织学观察. 结果 0. 25 CA组小鼠体重低于HFD组,总胆汁酸较HFD组升高,ALT水平、肝脏重量较HFD组升高(P<0. 05,P<0. 01),伴有脂滴数增多;UDCA组小鼠血糖水平较HFD组降低( P<0. 05 ) ,总胆汁酸水平、肝脏重量、脂滴数和血ALT水平与HFD组比较无明显变化(P>0. 05). 结论 对高脂诱导肥胖小鼠给予UDCA可降低其血糖水平,而胆酸可降低其体重,但应注意较高剂量胆酸可能导致的肝脏毒性.

목적 연구담즙산대고지유도비반소서혈당화체중적영향,병분석기안전성. 방법 32 지웅성C57BL/6소서,수궤분위고지음식조(HFD조)、고지음식가0. 1% w/w담산간예조(0. 1 CA조)、고지음식가0. 25%w/w담산간예조(0. 25 CA조)화고지음식가0. 25%웅거양담산간예조(UDCA조),매조8지. 간예후측정혈당、이도소、총담즙산、병안산전안매( ALT)화천동안산전안매( AST)적수평,채집간장조직칭중병진행조직학관찰. 결과 0. 25 CA조소서체중저우HFD조,총담즙산교HFD조승고,ALT수평、간장중량교HFD조승고(P<0. 05,P<0. 01),반유지적수증다;UDCA조소서혈당수평교HFD조강저( P<0. 05 ) ,총담즙산수평、간장중량、지적수화혈ALT수평여HFD조비교무명현변화(P>0. 05). 결론 대고지유도비반소서급여UDCA가강저기혈당수평,이담산가강저기체중,단응주의교고제량담산가능도치적간장독성.
Objective To investigate the effect of bile acid ( BA) on blood glucose and body weight of obese mice induced by high-fat diet and analyze its safety. Methods A total of 32 male C57BL/6 mice were randomly divided into high-fat diet group (HFD, n=8), HFD and 0. 1% w/w cholic acid (CA) intervention group (0. 1 CA group, n=8), HFD and 0. 25% w/w CA group (0. 25 CA group, n=8) and HFD and 0. 25% w/w Ursodeoxycholic Acid (UDCA group, n=8). After intervention, the levels of blood glucose, insulin, total bile acids, alanine transaminase (ALT) and aspartate aminotransferase ( AST) were detected, and liver tissues were weighed, and then histological observation was carried out. Results In 0. 25 CA group, the values of body weight were decreased, and the level of total bile acids was increased compared with that in HFD group (P<0. 05, P<0. 01), while values of ALT, liver weight and lipid droplet number were also increased. In UDCA group, blood glucose levels were decreased (P<0. 05, P<0. 01), while the differences in values of total bile acids, liver weight, lipid droplet number and serum were not statistically significant compared with those in HFD group (P>0. 05). Conclusion UDCA supplementation may reduce blood glucose level of obese mice induced by high-fat diet, and cholic acid supplementation may decrease weight gain. However, higher dose of cholic acid may result in hepatotoxicity.
