目的:探讨电针对胰岛素抵抗大鼠血清中超氧化物歧化酶( SOD)、丙二醛( MDA)及主动脉一氧化氮合酶( eNOS)蛋白表达的影响。方法以8只雄性LETO大鼠作为Blank组,将16只雄性OLETF大鼠随机分为E-A组及Model组,每组8只。 Blank组及Model组进行正常饲养,不给予其他处理。 E-A组针刺双侧内关、三阴交、足三里及肾俞,并予双侧足三里及三阴交加电。每日1次,每次20 min,持续4 w。治疗结束后,禁食12 h,次晨眼眶静脉窦采血。检测各组大鼠空腹血糖( FPG)、空腹胰岛素( FINS)、胰岛素敏感性指数( ISI)、SOD、MDA、一氧化氮( NO)及内皮素( ET)-1。 HE染色观察主动脉病理变化,Western印迹分析主动脉eNOS的蛋白表达。结果 Model组的FPG、FINS、MDA、ET相比Blank组显著升高(P=0.001,P=0.032,P=0.000,P=0.037),ISI、SOD、NO较Blank组显著降低(P=0.001,P=0.000,P=0.018)。 E-A组的 FPG、FINS、MDA较Model组显著降低(P=0.007,P=0.001,P=0.000),ISI、NO相比Model组显著升高(P=0.000,P=0.016),ET-1水平虽无统计学意义(P>0.05),但有所降低。光镜显示,电针干预对总体病理损伤有不同程度的改善。 E-A组eNOS蛋白表达明显高于Model组(P=0.005)。结论电针干预对胰岛素抵抗及氧化应激状态有良性调节作用,有助于抗氧化、抗血管损伤,促进内皮功能障碍的改善。

Objective To investigate the effect of electroacupuncture (E-A) on the serum levels of superoxide dismutase (SOD), malondialdehyde ( MDA) and the expression of endothelial nitric oxide synthase ( eNOS) of thoracic in OLETF rats .Methods Sixteen male OLETF rats were randomly divided into E-A and model groups ,eight rats in each group .Eight male LETO rats were divided as blank group . The E-A group was applied to bilateral Neiguan (PC6),Zusanli(ST36),Sanyinjiao(SP6),and Shenshu(BL23).Needles in Zusanli(ST36), Sanyinjiao(SP6) were connected with E-A.At the end of the experiment,glucose oxidase method,ELISA,WST-1,TBA and nitric acid deoxi-dize enzyme methods were used to detect the levels of fasting blood glucose (FPG),serum insulin (FINS),insulin sensitivity index (ISI), ET-1,SOD,MDA and NO.The aortic pathology was observed by HE staining .The expression of eNOS was detected by Western blot .Results Compared with those of blank group ,serum levels of FPG,FINS,MDA and ET-1 in model group were significantly increased (P=0.001, P=0.032,P=0.000,P=0.037),but the levels of ISI,SOD and NO were obviously decreased (P=0.001,P=0.000,P=0.018).Compared with those of model group,serum levels of FPG,FINS and MDA in E-A group were greatly decreased(P=0.007,P=0.001,P=0.000),the levels of ISI and NO were significantly increased (P=0.000,P=0.016).The level of ET-1 in E-A group was lower than that of model group even though it was no statistical significance (P>0.05).E-A improved morphological changes of thoracic .The expression of eNOS in E-A group was significantly higher than that of model group (P=0.005).Conclusions E-A intervention on insulin resistance and oxidative stress state has a positive adjustment which regulates oxidation resistance ,prevents blood vessel damage and promotes the improvement of endotheli -al dysfunction .
