目的:观察硫化氢( H2 S)对糖尿病大鼠心肌肌浆网钙泵( SERCA)活性及钙调控蛋白基因表达的影响。方法雄性SD大鼠随机分为正常对照组(NC组)、糖尿病对照组(DM组)、NaHS治疗组(NaHS+DM组)和NaHS对照组(NaHS组)。采用链脲佐菌素(STZ)55 mg/kg腹腔注射诱导糖尿病大鼠模型。测定大鼠的心系数;HE染色观察心肌组织的病理学变化;利用无机磷比色法测定大鼠心肌SERCA活性;RT-PCR检测心肌组织SERCA、受磷蛋白(PLB)、兰尼碱受体2型(RyR2)和1,4,5-三磷酸肌醇受体2型(IP3R2)的基因表达。结果与NC组相比,NaHS 组各项指标均无显著差异(P>0.05);而DM组心系数明显增大(P<0.01);心肌纤维排列紊乱,细胞肿胀明显,有炎细胞浸润;心肌 SERCA 的活性明显降低(P<0.01);SERCA、RyR2和IP3R2 mRNA表达降低(P<0.01),PLB mRNA表达增加(P<0.01)。与DM组相比,NaHS+DM组心系数明显降低(P<0.01),心肌病理学变化明显改善,SERCA活性明显增加(P<0.01),SERCA、RyR2和IP3R2 mRNA表达明显增加(P<0.01),PLB mRNA表达降低(P<0.01)。结论 H2 S对糖尿病大鼠心肌损伤具有保护作用,其机制可能与调节钙调控蛋白的基因表达,增强钙泵活性有关。

목적:관찰류화경( H2 S)대당뇨병대서심기기장망개빙( SERCA)활성급개조공단백기인표체적영향。방법웅성SD대서수궤분위정상대조조(NC조)、당뇨병대조조(DM조)、NaHS치료조(NaHS+DM조)화NaHS대조조(NaHS조)。채용련뇨좌균소(STZ)55 mg/kg복강주사유도당뇨병대서모형。측정대서적심계수;HE염색관찰심기조직적병이학변화;이용무궤린비색법측정대서심기SERCA활성;RT-PCR검측심기조직SERCA、수린단백(PLB)、란니감수체2형(RyR2)화1,4,5-삼린산기순수체2형(IP3R2)적기인표체。결과여NC조상비,NaHS 조각항지표균무현저차이(P>0.05);이DM조심계수명현증대(P<0.01);심기섬유배렬문란,세포종창명현,유염세포침윤;심기 SERCA 적활성명현강저(P<0.01);SERCA、RyR2화IP3R2 mRNA표체강저(P<0.01),PLB mRNA표체증가(P<0.01)。여DM조상비,NaHS+DM조심계수명현강저(P<0.01),심기병이학변화명현개선,SERCA활성명현증가(P<0.01),SERCA、RyR2화IP3R2 mRNA표체명현증가(P<0.01),PLB mRNA표체강저(P<0.01)。결론 H2 S대당뇨병대서심기손상구유보호작용,기궤제가능여조절개조공단백적기인표체,증강개빙활성유관。
Objective To investigate the effect of hydrogen sulfide (H2S) on sarcoplasmic reticulum Ca2+-ATPase (SERCA) activ-ity and gene expressions of calcium regulatory proteins in diabetic rats .Methods Thirty-two male SD rats were divided into four groups ran-domly:normal control (NC),diabetic (DM),NaHS+diabetic (NaHS+DM) and NaHS groups (NaHS).Diabetic rat was induced by injec-tion of streptozotocin at 55 mg/kg intraperitoneally .After 8 weeks,heart weight/body weight were determined .The pathological changes of myocardial tissue were observed by HE staining ,and the activity of SERCA was analyzed by colorimetric method of inorganic phosphorus .The expressions of SERCA,PLB (phospholamban),RyR2(ryanodine receptor type 2) and IP3R2 (1,4,5-trisphosphate inositor type 2) at mRNA level in myocardium were detected using RT-PCR.Results There were no significant differences in all indexes between NC and NaHS groups (P>0.05).However,compared with NC group,in diabetic rats,heart weight/body weight were increased (P<0.01).Cardiac muscle fibers were disorder ,and myocytes were edema and hypertrophy ,with inflammatory cell infiltration enhanced .The activity of SERCA was de-creased(P<0.01).The mRNA levels of SERCA,RyR2 and IP3R2 were decreased (P<0.01),PLB mRNA was increased (P<0.01) in DM group.Compared with DM group,heart weight/body weight was decreased (P<0.01) and the injury of myocardiac myofibrillae was attentuat-ed in NaHS+DM group.SERCA activity was increased (P<0.01).The mRNA levels of SERCA,RyR2 and IP3R2were increased (P<0.01),PLB mRNA was decreased (P<0.01).Conclusions H2S can protect myocardium in diabetic rats ,it is related to regulating the gene expressions of calcium regulatory proteins and enhancing the activity of sarcoplasmic reticulum Ca 2+-ATPase.
