目的:研究并探讨不同剂量下的柴胡水提组分对改良法致大鼠肝纤维化模型的保护作用。方法采用高脂、酒精刺激和猪血清等复合因素造成大鼠肝纤维化,造模同时给予不同剂量柴胡水提成分,并观察0、1、6、11、16、21天动物的一般情况、证候积分、体重、日食量、日水量,计算肝、脾、胸腺的脏器指数,检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、层黏连蛋白(LN)、血清透明质酸(HA)、III 型前胶原(PC-III)、IV 型胶原(IV-C)、生长转化因子-β1(TGF-β1)指标的水平。结果连续给予0.90~3.60 g·kg-1的柴胡水提成分,可改善大鼠饮食、饮水和活动情况,降低肝纤维化大鼠血清中 ALT、AST、LN、HA、PC-III、IV-C、TGF-β1水平,并呈现一定的剂量依赖性,脏器系数显著性降低。结论柴胡水提成分对改良法制大鼠纤维化具有明显的保护作用,且呈现良好的量效关系。
目的:研究併探討不同劑量下的柴鬍水提組分對改良法緻大鼠肝纖維化模型的保護作用。方法採用高脂、酒精刺激和豬血清等複閤因素造成大鼠肝纖維化,造模同時給予不同劑量柴鬍水提成分,併觀察0、1、6、11、16、21天動物的一般情況、證候積分、體重、日食量、日水量,計算肝、脾、胸腺的髒器指數,檢測血清中丙氨痠轉氨酶(ALT)、天鼕氨痠轉氨酶(AST)、層黏連蛋白(LN)、血清透明質痠(HA)、III 型前膠原(PC-III)、IV 型膠原(IV-C)、生長轉化因子-β1(TGF-β1)指標的水平。結果連續給予0.90~3.60 g·kg-1的柴鬍水提成分,可改善大鼠飲食、飲水和活動情況,降低肝纖維化大鼠血清中 ALT、AST、LN、HA、PC-III、IV-C、TGF-β1水平,併呈現一定的劑量依賴性,髒器繫數顯著性降低。結論柴鬍水提成分對改良法製大鼠纖維化具有明顯的保護作用,且呈現良好的量效關繫。
목적:연구병탐토불동제량하적시호수제조분대개량법치대서간섬유화모형적보호작용。방법채용고지、주정자격화저혈청등복합인소조성대서간섬유화,조모동시급여불동제량시호수제성분,병관찰0、1、6、11、16、21천동물적일반정황、증후적분、체중、일식량、일수량,계산간、비、흉선적장기지수,검측혈청중병안산전안매(ALT)、천동안산전안매(AST)、층점련단백(LN)、혈청투명질산(HA)、III 형전효원(PC-III)、IV 형효원(IV-C)、생장전화인자-β1(TGF-β1)지표적수평。결과련속급여0.90~3.60 g·kg-1적시호수제성분,가개선대서음식、음수화활동정황,강저간섬유화대서혈청중 ALT、AST、LN、HA、PC-III、IV-C、TGF-β1수평,병정현일정적제량의뢰성,장기계수현저성강저。결론시호수제성분대개량법제대서섬유화구유명현적보호작용,차정현량호적량효관계。
Objective To study the protective effect of different doses of aqueous extracted components of Bupleurum chinense against rats' liver fibrosis induced by composite factors. Methods The model of rats' liver fibrosis was established by the composite factors of high-fat diet, alcohol stimulation and porcine serum. Rats were observed in 0, 1, 6, 11, 16, 21 d in terms of animal general, syndrome integral, body weight, daily food intake, daily water, organ index of liver and spleen, thymus, calculation, serum the alanine aminotransferase (ALT), aspartate aminotransferase (AST), laminin (LN), serum hyaluronic acid (HA), procollagen type III (PC-III), type IV collagen (IV-C), transforming growth factor beta 1 (TGF- beta 1) index level. Results The diet, drinking water and physical activities of rats were improved by aqueous extracted components of Bupleurum chinense ranged from 0.90 g·kg-1 to 3.60g·kg-1 for 21 consecutive days by gavage. The contents of ALT, AST, HA, LN, IV-C and PC III and liver index declined significantly. Conclusion Liver fibrosis in rats induced by composite factors can be protected effectively by aqueous extracted components of Bupleurum chinense and shows a good dose-effect relationship.
