研究了以固定化的辣根过氧化酶为催化剂,催化H2O2氧化降解刚果红染料,考察了影响刚果红染料降解率的几个重要因素,包括pH﹑温度﹑反应时间﹑H2O2浓度﹑染料初始浓度等,确定其最佳条件为pH 3.5,固定化酶用量0.5 g,H2O2浓度1.0 mmol/L,底物浓度0.1 mmol/L,反应温度40℃,反应时间30 min,降解率达到91.6%. 催化剂重复使用4次后降解产率仍可达到22.0%,同时采用质谱对降解后的产物进行了分析.

Immobilized horseradish peroxidase was used as catalyst of oxidative degradation of Congo red dye by hydrogen per-oxide.Several important factors affecting the degradation rate of Congo red dye, including pH,temperature,reaction time, H2O2 concentration,the initial concentration of the dye etc.were investigated.The obtained optimum conditions are as follows:pH 3.5,0.5 g immobilized enzyme,1.0 mmol/L H2O2,0.1 mmol/L substrate concentration,40℃ reaction temperature and 30 min reac-tion time.Under the optimal conditions the degradation rate could reach to 91.6%.After use of catalyst four times,the rate of degradation rate can still reach to 22.0%.Furthermore,the degradation products were also analyzed by mass spectrometry.
