从广西宜州蚕区暴发黑胸败血病的养蚕农户处采集到病蚕样本,分离到1株病原菌,通过形态学观察和回复感染健康5龄家蚕试验及16S rDNA基因序列同源性分析鉴定,结果显示病原菌菌落呈橘黄色,光滑,圆形,边缘整齐,有光泽,湿润有黏性,革兰氏染色呈阳性,该菌形态学符合葡萄球菌属的特征;用菌液穿刺感染健康5龄家蚕10 h后蚕体胸部膨大,痉挛侧倒而死,11 h后胸部变黑呈现黑胸败血病病症;从腹腔接种菌液的小白鼠16 h内全部发病,从发病的家蚕体液和死亡小鼠的心血均能分离到与接种菌株形态一致的细菌, 证实该菌为引起家蚕黑胸败血病的病原菌. 菌株16S rDNA片段为1 463 bp (GenBank ID:1811543),与GenBank数据进行比较,该菌株与松鼠葡萄球菌(Staphylococcus sciuri)同源性为99.9%,鉴定该菌株为松鼠葡萄球菌.

A strain of pathogen was isolated from sick silworm which diagnosed cepticaemia in Yizhou.After morphological observation, reverse infection test on healthy five-year-old silkworm and 16s rDNA homology analysis, the results showed that the colony were orange, smooth, circular, neat, luster, wet sticky, and with Gram positive, the morphology of the bacterium was in line with characteristics of staphylococcus.Using bacteria liquid to puncture the infected five-year-old healthy silk-worm, its body chest enlarged 10 hours later and died with spasm.After 11 hours, its chest gradually turned into the symp-tom of cepticaemia.All the mice vaccinated the bacteria liquid from its vacuum were attacked of a disease in 16 hours, and a kind of germ with the same form of vaccinated bacteria was isolated both from the body fluid of sick silkworm and the heart blood and liver of the dead mouse which proved the pathogenic germs caused the silkworm cepticaemia.The 16s rDNA fragment of the strain was 1 463 bp (GeneBank ID: 1811543), and its homology with Staphylococcus sciuri was 99.9%.The bacteria germ strain was identified as Staphylococcus sciuri.
