从江西省宜春市温汤镇温泉水域中筛选出35株产α-淀粉酶嗜热菌株,选取一株产α-淀粉酶活力较高的嗜热菌YC-15为出发菌株,对其进行16S rRNA基因序列分析. 结果表明,YC-15与枯草芽孢杆菌(Bacillus subtilis)的16S rRNA序列相似性达98%,结合其形态﹑生理生化特征,进一步证明了YC-15在分类上应归属枯草芽孢杆菌.并对YC-15产酶条件进行了研究,其最佳碳源为2%可溶性淀粉,最佳氮源为0.5%酵母膏+0.5%蛋白胨,最适产酶初始pH为7.0,最适产酶温度55℃,属中性嗜热产α-淀粉酶菌.

35 thermophilic strains producing α-amylase were isolated from hot spring waters in Wentang town,Yichun city in Jiangxi .The strain which showed high α-amylase activity was selected and named as YC-15.The strain was identified as Bacillus subtilis based on analysis of 16S rRNA gene sequence which was exhibited 98% similarity with the 16S rRNA se-quences of Bacillus subtilis,as wall as morphological characteristics, physiological and biochemical characteristics.The optimum cultural conditions of strain YC-15 for the α-amylase production were as follows: The optimum carbon source for producingα-amylase was 2% soluble starch; the optimum nitrogen source was 0.5% yeast extract + 0.5% peptone ;and the optimum initial pH values were 7.0;and the optimum temperatures were 55℃, a neutral thermophiles strains producing α-amylase.
