目的:研究 Rho/ Rho 激酶的底物-肌球蛋白磷酸酶靶亚1(MYPT -1)及缝隙连接蛋白40(Cx40)在房颤患者左心耳中的表达,探讨 Rho/ Rho 激酶信号通路在房颤的发生和维持中的作用。方法40例接受开胸手术者分为房颤组和窦性心律组,手术中取约100 mg 左心耳组织。采用免疫组化 ABC 法、Western Blot 技术检测 MYPT -1、Cx40在左心耳组织的蛋白表达水平,并分析 MYPT -1、Cx40在房颤组中的相关性。结果免疫组化和 Western Blot 结果均显示,房颤组患者 MYPT -1、Cx40在左心耳中的表达明显高于窦性心律组( P ﹤0.01)。相关分析显示,房颤组 MYPT -1与Cx40的蛋白表达呈显著正相关( r =0.989,P ﹤0.01)。结论房颤患者左心耳组织中明显存在 MYPT -1表达上调与功能活化,提示 Rho 激酶信号转导通路在房颤病变中被激活,其机制可能因调节 Cx40而发挥作用。

목적:연구 Rho/ Rho 격매적저물-기구단백린산매파아1(MYPT -1)급봉극련접단백40(Cx40)재방전환자좌심이중적표체,탐토 Rho/ Rho 격매신호통로재방전적발생화유지중적작용。방법40례접수개흉수술자분위방전조화두성심률조,수술중취약100 mg 좌심이조직。채용면역조화 ABC 법、Western Blot 기술검측 MYPT -1、Cx40재좌심이조직적단백표체수평,병분석 MYPT -1、Cx40재방전조중적상관성。결과면역조화화 Western Blot 결과균현시,방전조환자 MYPT -1、Cx40재좌심이중적표체명현고우두성심률조( P ﹤0.01)。상관분석현시,방전조 MYPT -1여Cx40적단백표체정현저정상관( r =0.989,P ﹤0.01)。결론방전환자좌심이조직중명현존재 MYPT -1표체상조여공능활화,제시 Rho 격매신호전도통로재방전병변중피격활,기궤제가능인조절 Cx40이발휘작용。
Objective To explore the expressions of myosin phosphatase target subunit - 1(MYPT - 1)and connexin 40(Cx40)in left atrial appendage of patients with atrial fibrillation(AF). Methods Left atrial appendage tissue samples of 40 patients undergoing cardiac surgery were examined. Among the patients examined,20 had AF,20 had no history of AF. The expressions of MYPT - 1 and Cx40 in left atrial append-age were detected by immunohistochemistry and western blot. Results Immunohistochemistry and western blot showed that the expressions of MYPT - 1 and Cx40 were significantly up - regulated in left atrial appendage of patients with AF than patients with sinus rhythm( P ﹤ 0. 01). The two indicators were positively correlated( r = 0. 989,P ﹤ 0. 01). Conclusion The MYPT - 1 expression in left atrial appendage of patients with AF is up - regulated and activated,MYPT - 1 promotes the expression of Cx40,which suggests that Rho kinase signal transduction may play an important role in the pathological changes in AF.
