目的:研究大蒜素干预下 SD 大鼠血清对食管癌 EC109细胞增殖活性的影响。方法采用普通 SD 大鼠血清(Ⅰ组)和长期大蒜素干预条件下 SD 大鼠血清(Ⅱ组)代替胎牛血清培养食管癌 EC109细胞;用 MTT 比色法检测吸光度值,并根据吸光度值计算出细胞生长抑制率;应用流式细胞术测定细胞周期分布,并计算细胞 S 期分数和增殖指数。结果在相同的时间点上Ⅱ组细胞生长抑制率强于Ⅰ组,差异统计学意义( P ﹤0.05)。Ⅱ组 G2/ M 期及 S 期细胞比率明显低于Ⅰ组,而 G0/ G1期细胞比率明显升高;Ⅱ组 S 期分数( t =3.698)及细胞增殖指数( t =2.572)显著低于Ⅰ组,差异有统计学意义( P ﹤0.05)。结论长期大蒜素干预条件下 SD 大鼠血清能抑制食管癌 EC109细胞的生长以及DNA 合成。大蒜对食管癌 EC109细胞的增殖活性具有明显的抑制作用。

목적:연구대산소간예하 SD 대서혈청대식관암 EC109세포증식활성적영향。방법채용보통 SD 대서혈청(Ⅰ조)화장기대산소간예조건하 SD 대서혈청(Ⅱ조)대체태우혈청배양식관암 EC109세포;용 MTT 비색법검측흡광도치,병근거흡광도치계산출세포생장억제솔;응용류식세포술측정세포주기분포,병계산세포 S 기분수화증식지수。결과재상동적시간점상Ⅱ조세포생장억제솔강우Ⅰ조,차이통계학의의( P ﹤0.05)。Ⅱ조 G2/ M 기급 S 기세포비솔명현저우Ⅰ조,이 G0/ G1기세포비솔명현승고;Ⅱ조 S 기분수( t =3.698)급세포증식지수( t =2.572)현저저우Ⅰ조,차이유통계학의의( P ﹤0.05)。결론장기대산소간예조건하 SD 대서혈청능억제식관암 EC109세포적생장이급DNA 합성。대산대식관암 EC109세포적증식활성구유명현적억제작용。
Objective To study the inhibitory effect of allicin - intervened SD rat,serum on esophagus cancer EC - 109 cell proliferation activity. Methods The common SD rats,serum(groupⅠ)and the SD rats,serum under the condition of long - term allicin intervention(groupⅡ)instead of fetal calf serum were used respectively to cultivate esophagus cancer EC - 109 cells. MTT colorimetric method was used to measure the absorbance value,and according to the value to calculate the cell growth inhibition rate. Flow cytometry was used to determine cell cycle distri-bution,S - phase fraction and proliferation index. Results At the same time point,the inhibition rate of group Ⅱ was higher than groupⅠ,and there was a significant statistical difference( P ﹤ 0. 05). The proportion of G2 / M phase and S phase was much lower in group Ⅱthan groupⅠ, but the G0 / G1 phase cell proportion was much higher. The S - phase fraction( t = 3. 698)and proliferation index( t = 2. 572)of the group Ⅱwere much lower than group Ⅰ( P ﹤ 0. 05)and the difference was statistically significant( P ﹤ 0. 05). Conclusion The SD rat,serum under the condition of long - term allicin intervention can inhibit the growth and DNA synthesis of EC - 109 cell. The allicin has significant inhibitory effect on the proliferative activity of EC - 109 cell line.
