建立一种简单、准确度高、适用性强的方法,用于检测含植物提取物固体饮料中总黄酮的含量.对样品处理的提取溶剂浓度、超声时间、提取料液比进行优化,并对该测定方法低浓度范围内的标准曲线及测定方法的精密度和准确度进行分析.样品最佳处理条件为:提取溶剂为70%乙醇,超声时间30 min,提取料液比1:2.5(g/mL),该方法的RSD在1.03%~7.83%范围内,精确度较好,平均加样回收率为98.44%,标准曲线在低浓度0~20μg/mL范围内的线性方程为Y=0.012 7X-0.002 4,R=0.999 3,线性关系良好.该法操作简单,对于含植物提取物固体饮料中总黄酮的测定适用性好,准确度高,可为固体饮料中总黄酮测定方法标准的制定提供参考.

A more applicable and accurate method was developed to determine the total flavonoids in solid drink with plant extract by spectrophotometric method in this study. The effects of the concentration of solvent , ultrasonic time and ratio of solid to liquid on the determination of total flavonoids were studied. The standard curve in low rutin concentration and the precision and accuracy of that method were analyzed. The optimum conditions were as follow:the concentration of solvent was 70%ethanol , the ultrasonic time was 30 min and the ratio of solid to liquid was 1:2.5 (g/mL). Under this condition, this method showed good linearity (Y=0.012 7X-0.002 4, R=0.999 3) in low rutin concentration (0-20μg/mL) and average recovery (98.44%) as well as low RSD (1.03 %-7.83 %). This simple method was regarded to be very useful and high accuracy for the determination of total flavanoids in solid drink with plant extract.
