BUSHENJIANGUTANG%Periodontitis%Rats
目的:通过检测牙周炎大鼠血清中白细胞介素-4(interleukin-4,IL-4)、白细胞介素-6(interleukin-6, IL-6)和肿瘤坏死因子-α(tumor necrosis factor-α, TNF-α)水平的变化,及牙周组织的形态改变,初步探讨补肾坚骨汤对牙周炎大鼠的治疗作用。方法雄性12月龄SD大鼠45只,其中8只为正常对照组,37只采用丝线结扎8周,建立大鼠牙周炎模型后,将建模成功大鼠(34只)随机分为牙周炎模型对照组10只,补肾坚骨汤高剂量组(16g/kg/d)8只,补肾坚骨汤中剂量组(8g/kg/d)8只,和补肾坚骨汤低剂量组(4g/kg/d)8只,给予补肾坚骨汤灌胃4周。各组大鼠末次给药后24 h处死,采用ELISA法检测外周血IL-4、IL-6和TNF-α的水平;取上颌骨制作牙体牙周组织联合切片,HE染色,光学显微镜下观察牙周的组织学改变。结果牙周炎模型对照组血清IL-6、TNF-α水平显著高于正常对照组,IL-4水平显著低于正常对照组。补肾坚骨汤各剂量治疗组血清IL-6、TNF-α水平显著低于牙周炎模型对照组,补肾坚骨汤高、中剂量治疗组血清IL-4水平显著高于牙周炎模型对照组(p<0.05)。牙周炎模型对照组牙周组织表现为明显的炎症破坏,牙槽骨吸收明显,而补肾坚骨汤高、中剂量治疗组牙周组织炎症均明显减轻,牙槽骨吸收减少且骨修复反应明显。结论补肾坚骨汤可能通过降低牙周炎大鼠血清中IL-6和TNF-α水平,升高IL-4水平,减轻牙周组织炎症,减缓牙周组织破坏。

목적:통과검측아주염대서혈청중백세포개소-4(interleukin-4,IL-4)、백세포개소-6(interleukin-6, IL-6)화종류배사인자-α(tumor necrosis factor-α, TNF-α)수평적변화,급아주조직적형태개변,초보탐토보신견골탕대아주염대서적치료작용。방법웅성12월령SD대서45지,기중8지위정상대조조,37지채용사선결찰8주,건립대서아주염모형후,장건모성공대서(34지)수궤분위아주염모형대조조10지,보신견골탕고제량조(16g/kg/d)8지,보신견골탕중제량조(8g/kg/d)8지,화보신견골탕저제량조(4g/kg/d)8지,급여보신견골탕관위4주。각조대서말차급약후24 h처사,채용ELISA법검측외주혈IL-4、IL-6화TNF-α적수평;취상합골제작아체아주조직연합절편,HE염색,광학현미경하관찰아주적조직학개변。결과아주염모형대조조혈청IL-6、TNF-α수평현저고우정상대조조,IL-4수평현저저우정상대조조。보신견골탕각제량치료조혈청IL-6、TNF-α수평현저저우아주염모형대조조,보신견골탕고、중제량치료조혈청IL-4수평현저고우아주염모형대조조(p<0.05)。아주염모형대조조아주조직표현위명현적염증파배,아조골흡수명현,이보신견골탕고、중제량치료조아주조직염증균명현감경,아조골흡수감소차골수복반응명현。결론보신견골탕가능통과강저아주염대서혈청중IL-6화TNF-α수평,승고IL-4수평,감경아주조직염증,감완아주조직파배。
Objective To investigate the effect of BUSHENJIANGUTANG on the serum levels of tumor necrosis factor-α (TNF-α), interleukin-4(IL-4)and interleukin-6(IL-6) and to examine the morphological changes of periodontal tissue in ligature-induced periodontitis rats. Methods Forty-five 12-month-old male SD rats were divided into control group (8 rats), and periodontitis groups (37 rats)in which periodontitis was induced by ligature on the maxillary second molars. At 8 weeks after ligature, the periodontitis model rats were successfully established(34 rats) and divided into four groups, periodontitis rats without treatment ( 10 rats ), periodontitis rats with 16g/kg/d BUSHENJIANGUTANG intragastrical administration(8 rats), periodontitis rats with 8g/kg/d BUSHENJIANGUTANG intragastrical administration (8 rat), and periodontitis rats with 4g/kg/d BUSHENJIANGUTANG intragastrical administration(8 rats) for four weeks. The rats were sacrificed after the last administration. The histology of the periodontium was examined and serum IL-4,IL-6 and TNF-α were detected by ELISA. Results Serum IL-6 and TNF-αwere significantly higher and serum IL-4 was lower in periodontitis rats without treatment than those in normal control group. Serum IL-6 and TNF-α in BUSHENJIANGUTANG intragastrical administration group were significantly lower than those in periodontitis rats without treatment group, serum IL-4 in periodontitis rats with 16g/kg/d and 8g/kg/d BUSHENJIANGUTANG administration group was significantly higher than that in periodontitis rats without treatment group ( p<0. 05 ) . There was obvious inflammation and alveolar bone loss in periodontitis rats without treatment group. BUSHENJIANGUTANG intragastrical administration rats showed less inflammations and less bone loss. Conclusion BUSHENJIANGUTANG may reduce the inflammation of periodontal tissues and slow the destruction of periodontal tissues by decreasing the serum TNF- and IL-6 levels and increasing IL-4 level in rats with periodontitis.
