目的：繁殖和鉴定烟碱型乙酰胆碱受体α7基因敲除小鼠。方法：将引进的α7nAChR基因敲除杂合子小鼠进行饲养并繁殖，提取子代小鼠尾部DNA，用PCR方法检测子代小鼠基因型；采用 Western blot 检测α7nAChR 蛋白质的表达，进一步证实PCR鉴定方法的可靠性。结果：获得子代（α7nAChR＋／＋），子代小鼠纯合率为30％。结论：α7nAChR基因敲除小鼠的繁育和鉴定均获得成功，为进一步研究α7nAChR缺失对非酒精性脂肪性肝炎的影响提供了理想的动物模型。
目的：繁殖和鑒定煙堿型乙酰膽堿受體α7基因敲除小鼠。方法：將引進的α7nAChR基因敲除雜閤子小鼠進行飼養併繁殖，提取子代小鼠尾部DNA，用PCR方法檢測子代小鼠基因型；採用 Western blot 檢測α7nAChR 蛋白質的錶達，進一步證實PCR鑒定方法的可靠性。結果：穫得子代（α7nAChR＋／＋），子代小鼠純閤率為30％。結論：α7nAChR基因敲除小鼠的繁育和鑒定均穫得成功，為進一步研究α7nAChR缺失對非酒精性脂肪性肝炎的影響提供瞭理想的動物模型。
목적：번식화감정연감형을선담감수체α7기인고제소서。방법：장인진적α7nAChR기인고제잡합자소서진행사양병번식，제취자대소서미부DNA，용PCR방법검측자대소서기인형；채용 Western blot 검측α7nAChR 단백질적표체，진일보증실PCR감정방법적가고성。결과：획득자대（α7nAChR＋／＋），자대소서순합솔위30％。결론：α7nAChR기인고제소서적번육화감정균획득성공，위진일보연구α7nAChR결실대비주정성지방성간염적영향제공료이상적동물모형。
Objective:Reproduce and identify α7nAChR knockout mice.Methods:Heterozygote mice were bred and reproduced.Ge-nome DNA was extracted from the murine tail and identified by PCR;Additionally, the expression ofα7nAChR protein was detected by Western blot to further confirmed the reliability of PCR methods for genotype identification of α7nAChR mutant mice.Results:α7nAChR genotypes of the offspring mice included heterozygous (α7nAChR+/-) ,homozygous (α7nAChR-/-) ,and wild types (α7nAChR+/+) .The homozygous rate of offspring was 30% .Conclusion: α7nAChR knockout mice are successful reproduced, which provides an ideal animal model for study of the NASH.
