以 GenBank中登载的普通猪FABP4基因为参考序列,设计特异性引物,采用 RT-PCR技术从版纳微型猪近交系卵巢组织中扩增FABP4基因的编码区序列,经直接测序后采用生物信息学软件对编码区序列和蛋白质结构进行生物信息学分析和结构预测。结果表明:版纳微型猪近交系(BMI)FABP4基因编码区序列长399 bp,编码132个氨基酸;BMI FABP4基因同普通猪比对同源性为100%,未发现碱基突变;生物信息学分析表明不同物种FABP4基因编码区核苷酸序列与氨基酸序列具有较高的保守性;蛋白结构预测显示,FABP4蛋白空间结构为由2个α螺旋和10个反向平行的β折叠围成的桶状结构。

The CDS region and partial sequence of fatty acid binding protein 4 (FABP4)gene of Banna Mini-pig Inbred Line (BMI)were amplified using RT-PCR with specific primers that designed according to cDNA sequence of swine FABP4 gene in the GenBank so as to analyze the bioinformatics of putative pro-tein.The result showed that the sequence length of coding region of the BMI FABP4 gene was 3 9 9 bp,en-coding a protein of 132 amino acids.The homology of FABP4 gene between BMI and swine was 100% and no base mutation was found.By biology software analysis,the nucleotide sequence of the coding region and accordingly amino acid sequences of FABP4 gene of different species were conserved in evolution.The FABP4 protein was assembled as a barrel conformation by twoαhelixes and tenβfolds.
