目的 探讨骨癌痛(bone cancer pain,BCP)大鼠脊髓背角酸敏感离子通道1a (acid-sensing ion channel 1a,ASIC1a)表达的变化. 方法 雌性未交配SD大鼠56只,体重180 g~220 g,采用随机数字表法分为3组:正常组(Ⅰ组,12只)、假手术组(Ⅱ组,12只)和BCP组(Ⅲ组,32只).每组随机取8只分别于术前1d及术后3、5、7、10、14、18d测定大鼠体重和机械缩足反射阈值(paw withdrawal mechanical threshold,PWMT).Ⅰ组、Ⅱ组于术后18d测完PWMT后,Ⅲ组于术前1d及术后3、7、10、14、18d,同一时段随机处死4只大鼠,取腰段脊髓,采用Western blot法检测ASIC1a的表达.每组于术后14 d取4只大鼠,采用免疫荧光技术观察各组大鼠患侧脊髓ASIC1a和神经元免疫反应阳性产物的共表达. 结果 与Ⅰ组、Ⅱ组比较,Ⅲ组术后7d~18d体重降低(P<0.01);5 d PWMT[(8.86±2.49)g]开始下降(P<0.01),术后7[(5.17±1.26)g]、10 [(2.68±0.60)g]、14[(1.85±0.98)g]、18 d[(1.58±0.38) g]PWMT进一步降低(P<0.01);Western blot显示,Ⅲ组术后3d开始ASIC1a表达上调(P<0.01),于术后14 d~18 d达到高峰(P<0.01);免疫荧光发现,Ⅲ组脊髓背角ASIC1a的表达显著增加(P<0.01),与神经元共标细胞数也相应增加(P<0.01). 结论 BCP大鼠脊髓背角神经元中ASIC1a表达增多,大鼠BCP的形成和维持可能与脊髓背角ASIC1a表达上调有关.

목적 탐토골암통(bone cancer pain,BCP)대서척수배각산민감리자통도1a (acid-sensing ion channel 1a,ASIC1a)표체적변화. 방법 자성미교배SD대서56지,체중180 g~220 g,채용수궤수자표법분위3조:정상조(Ⅰ조,12지)、가수술조(Ⅱ조,12지)화BCP조(Ⅲ조,32지).매조수궤취8지분별우술전1d급술후3、5、7、10、14、18d측정대서체중화궤계축족반사역치(paw withdrawal mechanical threshold,PWMT).Ⅰ조、Ⅱ조우술후18d측완PWMT후,Ⅲ조우술전1d급술후3、7、10、14、18d,동일시단수궤처사4지대서,취요단척수,채용Western blot법검측ASIC1a적표체.매조우술후14 d취4지대서,채용면역형광기술관찰각조대서환측척수ASIC1a화신경원면역반응양성산물적공표체. 결과 여Ⅰ조、Ⅱ조비교,Ⅲ조술후7d~18d체중강저(P<0.01);5 d PWMT[(8.86±2.49)g]개시하강(P<0.01),술후7[(5.17±1.26)g]、10 [(2.68±0.60)g]、14[(1.85±0.98)g]、18 d[(1.58±0.38) g]PWMT진일보강저(P<0.01);Western blot현시,Ⅲ조술후3d개시ASIC1a표체상조(P<0.01),우술후14 d~18 d체도고봉(P<0.01);면역형광발현,Ⅲ조척수배각ASIC1a적표체현저증가(P<0.01),여신경원공표세포수야상응증가(P<0.01). 결론 BCP대서척수배각신경원중ASIC1a표체증다,대서BCP적형성화유지가능여척수배각ASIC1a표체상조유관.
Objective To assess the role of acid-sensing ion channel 1a (ASIC1a) in the dorsal horn of spinal cord in a rat model of bone cancer pain(BCP).Methods Fifty six female SD rats weighting 180 g-220 g were randomly divided into 3 groups: normal group (group Ⅰ , n=12), sham operation group (group Ⅱ , n=12), and bone cancer pain group (group Ⅲ, n=32).Body weight and paw withdrawal mechanical threshold(PWMT) of mechanical stimulus with von-Frey filaments were determined on the day before operation and on the days 3, 5, 7, 10, 14 d and 18 d after operation.Four animals were killed randomly on the 18 d after operation in groups Ⅰ and Ⅱ and at 3, 7, 10, 14 d and 18 d after operation in group Ⅲ.The lumbar segments of the spinal cord were removed to determine expression of ASIC1a by Western blot.Four animals were sacrificed on the 14 d after operation and the L4-6spinal cord segments were removed to measure the expressions of ASIC1a and the colocalization of ASIC 1a and neuronal nuclei in the dorsal horn by immunofluorescence staining.Results The PWMT was significantly decreased on the 5 [(8.86±2.49) g], 7 [(5.17±1.26) g], 10 [(1.85±0.98) g], 18 d [(1.58±0.38) g] after operation (P<0.01), whereas body weight difference was increased in the group Ⅲ on the 7 to 18 days compared with groups Ⅰ and Ⅱ (P<0.01).Compared with groups Ⅰ and Ⅱ , spinal expression of ASIC1a in the group Ⅲ was significantly increased from 3 d to 18 d after operation (P<0.01), and reached peak on the 14 d to 18 d after operation (P<0.01).The immunofluorescence staining showed increased-ASIC1a concentrated in the superficial laminae (Ⅰ and Ⅱ) in the spinal dorsal horn ipsilateral to hind paw of bone cancer pain rats (P<0.01).Double staining showed that ASIC1a was colocalized with neuronal nuclei and the number was increased on the 14 d after operation (P<0.01).Conclusions Expression of ASIC1a was up-regulated in the superficial dorsal horn neurons of spinal cord in BCP rats.ASIC1a in the spinal cord may be involved in the mechanism of BCP.
