目的 观察心力衰竭( HF)大鼠血浆N端脑钠肽前体( NT-proBNP)水平及CD4+CD2+5 Foxp3+Treg细胞比例变化,评价脐带间充质干细胞( WJ-MSCs)移植对HF大鼠免疫激活状态的影响. 方法 雄性Wistar大鼠48只,空白组(16只)不作处理正常饲养;其余大鼠皮下注射异丙肾上腺素制造HF模型成功后,随机分为干细胞移植组(16只)和模型组( 16只). 干细胞移植组心肌内注射WJ-MSCs悬液,模型组予心肌内注射等量生理盐水. 4周用流式细胞技术测定外周血CD4+CD2+5 Foxp3+Treg细胞比例,酶联免疫吸附法测定三组大鼠血清中NT-proBNP及TNF-α、IL-10水平,并取大鼠心脏组织行免疫组化染色. 结果 HF大鼠血浆NT-proBNP、TNF-α均高于空白组,外周血CD4+CD2+5Foxp3+Treg细胞比例及IL-10水平低于空白组(P<0.05);移植后4周,与模型组比较干细胞移植组NT-proBNP、TNF-α水平明显降低,外周血CD4+CD2+5 Foxp3+Treg细胞比例及IL-10 水平升高( P均 <0.05 );CD4+CD2+5 Foxp3+Treg细胞比例与血浆NT-proBNP水平呈负相关(r=-0.634,P<0.05). 结论 WJ-MSCs移植可显著提高HF大鼠外周血CD4+CD2+5 Foxp3+Treg细胞比例,抑制炎症反应,改善HF症状.
目的 觀察心力衰竭( HF)大鼠血漿N耑腦鈉肽前體( NT-proBNP)水平及CD4+CD2+5 Foxp3+Treg細胞比例變化,評價臍帶間充質榦細胞( WJ-MSCs)移植對HF大鼠免疫激活狀態的影響. 方法 雄性Wistar大鼠48隻,空白組(16隻)不作處理正常飼養;其餘大鼠皮下註射異丙腎上腺素製造HF模型成功後,隨機分為榦細胞移植組(16隻)和模型組( 16隻). 榦細胞移植組心肌內註射WJ-MSCs懸液,模型組予心肌內註射等量生理鹽水. 4週用流式細胞技術測定外週血CD4+CD2+5 Foxp3+Treg細胞比例,酶聯免疫吸附法測定三組大鼠血清中NT-proBNP及TNF-α、IL-10水平,併取大鼠心髒組織行免疫組化染色. 結果 HF大鼠血漿NT-proBNP、TNF-α均高于空白組,外週血CD4+CD2+5Foxp3+Treg細胞比例及IL-10水平低于空白組(P<0.05);移植後4週,與模型組比較榦細胞移植組NT-proBNP、TNF-α水平明顯降低,外週血CD4+CD2+5 Foxp3+Treg細胞比例及IL-10 水平升高( P均 <0.05 );CD4+CD2+5 Foxp3+Treg細胞比例與血漿NT-proBNP水平呈負相關(r=-0.634,P<0.05). 結論 WJ-MSCs移植可顯著提高HF大鼠外週血CD4+CD2+5 Foxp3+Treg細胞比例,抑製炎癥反應,改善HF癥狀.
목적 관찰심력쇠갈( HF)대서혈장N단뇌납태전체( NT-proBNP)수평급CD4+CD2+5 Foxp3+Treg세포비례변화,평개제대간충질간세포( WJ-MSCs)이식대HF대서면역격활상태적영향. 방법 웅성Wistar대서48지,공백조(16지)불작처리정상사양;기여대서피하주사이병신상선소제조HF모형성공후,수궤분위간세포이식조(16지)화모형조( 16지). 간세포이식조심기내주사WJ-MSCs현액,모형조여심기내주사등량생리염수. 4주용류식세포기술측정외주혈CD4+CD2+5 Foxp3+Treg세포비례,매련면역흡부법측정삼조대서혈청중NT-proBNP급TNF-α、IL-10수평,병취대서심장조직행면역조화염색. 결과 HF대서혈장NT-proBNP、TNF-α균고우공백조,외주혈CD4+CD2+5Foxp3+Treg세포비례급IL-10수평저우공백조(P<0.05);이식후4주,여모형조비교간세포이식조NT-proBNP、TNF-α수평명현강저,외주혈CD4+CD2+5 Foxp3+Treg세포비례급IL-10 수평승고( P균 <0.05 );CD4+CD2+5 Foxp3+Treg세포비례여혈장NT-proBNP수평정부상관(r=-0.634,P<0.05). 결론 WJ-MSCs이식가현저제고HF대서외주혈CD4+CD2+5 Foxp3+Treg세포비례,억제염증반응,개선HF증상.
Objective To observe the levels of N-terminal pro-brain natriuretic peptide ( NT-proBNP) and the per-centage change of CD4+CD2+5 Foxp3+Treg cells in rats with heart failure ( HF) and to evaluate the effect of Wharton's jelly mesenchymal stem cells ( WJ-MSCs) transplantation on the immune activation of HF rats.Methods Forty-eight male Wistar rats were selected and 16 rats were fed normally without any treatment which was taken as the blank group, the rest of the rats were subcutaneously injected with isoprenaline to make the HF models, after the success of modeling, they were randomly divided into the stem cell transplantation group (n=16) and the model group (n=16).The rats in the stem cell transplantation group was administered with intramyocardial injection of WJ-MSCs suspension, while the rats in the model group was injected with the same amount of normal saline.Four weeks later, CD4+CD2+5 Foxp3+Treg cell proportion of the peripheral blood was detected by flow cytometry, the levels of NT-proBNP, TNF-αand IL-10 in the serum of the three groups were detected by enzyme-linked immunosorbent assay ( ELISA) , and the heart tissues of rats were collected for the immunohistochemical staining.Results The levels of NT-proBNP and TNF-αin the plasma of HF rats were significantly higher than those of the blank group, the CD4+CD2+5 Foxp3+Treg cell proportion in the peripheral blood and the level of IL-10 was significantly lower than that of the blank group (P<0.05).Four weeks after cell transplantation, compared with the model group, NT-proBNP and TNF-αwere significantly decreased, while the CD4+CD2+5 Foxp3+Treg cell proportion in the peripheral blood and the IL-10 level were significantly increased in the stem cell transplantation group, and the differ-ence was statistically significant (all P<0.05).There was negative correlation between the level of CD4+CD2+5 Foxp3+Treg cell and NT-proBNP expression (r=-0.634,P<0.05).Conclusion WJ-MSCs transplantation can significantly in-crease the CD4+CD2+5 Foxp3+Treg cell proportion in the peripheral blood of HF rats, inhibit inflammatory reaction and im-prove the symptoms of HF.
