目的 观察大鼠肝缺血再灌注损伤模型肺组织的氧化应激状态以及抗氧化酶过氧化氢酶(CAT)、超氧化物歧化酶(SOD)的表达变化,探讨肝缺血再灌注损伤对肺组织的影响和CAT、SOD的抗氧化作用.方法 采用无损伤血管夹夹闭通往大鼠肝右叶、肝中叶的血管和胆管30 min,然后松开血管夹恢复肝脏血液供应,制造大鼠70%肝脏缺血再灌注损伤模型;再灌注6h后取血、肝脏和肺.血清丙氨酸氨基转移酶(ALT)活性采用速率法测定.采用HE染色法观察肝脏的形态学改变.肺组织丙二醛(MDA)含量采用硫代巴比妥酸比色法测定,抗氧化酶CAT、SOD的mRNA表达水平采用RT-PCR的方法测定,蛋白水平的表达变化采用Western blot法测定.结果 与对照组相比,肝脏缺血再灌注损伤组大鼠血清ALT活性和肺组织内MDA的含量均明显升高,HE染色结果显示肝组织结构受损明显,肺组织抗氧化酶CAT、SOD的mRNA水平和蛋白水平也均明显升高.结论 肝缺血再灌注损伤使肺组织也遭受过氧化损伤;抗氧化酶CAT、SOD在此过程中可能发挥了抗氧化应激作用,对肺脏具有保护作用.

Objective To observe the oxidative stress level of lung tissue and expression change of catalase (CAT) and superoxide dismutase (SOD) in hepatic ischemia-reperfusion injury (HIRI) model of rats, to explore the effect of HIRI on lung tissue and antioxidant function of CAT and SOD.Methods The HIRI model of rats was established by clipping the hepatic blood vessel both hepatic right and middle lobes as well as bile duct with non-damage vascular clamp for 30 mins.The serum, hepatic tissues and lung were taken followed by six hours clipping.The activity of serum alanine aminotransferase (ALT) was detected with rate method.The morphological changes of liver were observed with HE staining.The malonaldehyde (MDA) content in lung tissue was determined with thiobarbituric acid colorimetric method.The expressions of CAT and SOD mRNA in lung tissue were evaluated by RT-PCR and protein levels of CAT and SOD were estimated by Western blot.Results Compared with the control group,the activity of ALT in serum,MDA content in lung tissue of HIRI rats were significantly increased.Results of HE staining showed that structure of liver tissue of HIRI model was significantly impaired.The mRNA and protein levels of CAT and SOD in lung tissue of HIRI model were also increased significantly.Conclusions HIRI can also cause the peroxidative damage of lung tissue.The antioxidase CAT and SOD may play an important role in inhibiting the oxidative-stress response during the process and have the protecting function for lung tissue.
