DNA双链断裂(DNA double strand breaks,DSBs)是诸多DNA损伤类型中对基因组的完整性、稳定性影响最严重的一种损伤形式.选择性非同源末端连接(alternative nonhomologous end joining,A-NHEJ)是最近研究新发现的一种不依赖Ku蛋白的DSBs修复途径,由于它是一种易错的修复方式,因此能引起基因组不稳定甚至肿瘤发生.本文综述了A-NHEJ的发现以及其修复过程和特点,并介绍了A-NHEJ在染色体易位、端粒融合、V(D)J重排和类别转换重组中的DSBs修复作用.
DNA雙鏈斷裂(DNA double strand breaks,DSBs)是諸多DNA損傷類型中對基因組的完整性、穩定性影響最嚴重的一種損傷形式.選擇性非同源末耑連接(alternative nonhomologous end joining,A-NHEJ)是最近研究新髮現的一種不依賴Ku蛋白的DSBs脩複途徑,由于它是一種易錯的脩複方式,因此能引起基因組不穩定甚至腫瘤髮生.本文綜述瞭A-NHEJ的髮現以及其脩複過程和特點,併介紹瞭A-NHEJ在染色體易位、耑粒融閤、V(D)J重排和類彆轉換重組中的DSBs脩複作用.

DNA double-strand breaks (DSBs) are the most severe damage of many types of DNA lesions that affect the genomic integrity and stability.Recently, a new Ku-independent DSBs repair pathway, alternative non-homologous end joining (A-NHEJ) , has been discovered, which is an errorprone repair pathway and thus may lead to genomic instability and even tumorigenesis.This review summarizes the discovery, repair process and characteristics of A-NHEJ, and also introduces theDSBs repair role of A-NHEJ in the process of chromosomal translocation, telomere fusion, V(D)J rearrangement and class switch recombination.
