目的:探讨益气活血方对冠心病心气虚大鼠模型心肌细胞凋亡的影响,以揭示冠心病心气虚证发病的病理改变,为中药治疗冠心病心气虚证提供实验依据。方法:采用“左冠状动脉结扎术加升压药”法制备冠心病心气虚大鼠模型,并将大鼠分为空白组、模型组、假手术组,益气活血方高、中、低剂量组,西药组(盐酸地尔硫)。于造模45 d后进行麻醉,取心肌组织,HE染色检测结扎处心肌细胞形态学变化。结果:与空白组比较,模型组心肌细胞排列紊乱,细胞坏死,较多炎性细胞浸润,细胞核固缩变形甚至脱失,血管管腔增大,组织间隙明显增宽并充填有大量纤维组织,横纹模糊断裂;与模型组比较,益气活血方高剂量组细胞凋亡改善明显,心肌细胞排列较为整齐,未见明显炎性细胞浸润,无明显血管扩张,纤维增生程度减轻。结论:中药益气活血方能够抑制并扭转心肌细胞的凋亡而减轻心肌缺血缺氧导致的细胞损伤。

목적:탐토익기활혈방대관심병심기허대서모형심기세포조망적영향,이게시관심병심기허증발병적병리개변,위중약치료관심병심기허증제공실험의거。방법:채용“좌관상동맥결찰술가승압약”법제비관심병심기허대서모형,병장대서분위공백조、모형조、가수술조,익기활혈방고、중、저제량조,서약조(염산지이류)。우조모45 d후진행마취,취심기조직,HE염색검측결찰처심기세포형태학변화。결과:여공백조비교,모형조심기세포배렬문란,세포배사,교다염성세포침윤,세포핵고축변형심지탈실,혈관관강증대,조직간극명현증관병충전유대량섬유조직,횡문모호단렬;여모형조비교,익기활혈방고제량조세포조망개선명현,심기세포배렬교위정제,미견명현염성세포침윤,무명현혈관확장,섬유증생정도감경。결론:중약익기활혈방능구억제병뉴전심기세포적조망이감경심기결혈결양도치적세포손상。
Objective:Investigating the effect of Yiqi Huoxue decoction on myocardium cell apoptosis in rats model with coronary heart disease (CHD)(heart qi deficiency) so as to reveal the pathologic change of CHD (heart qi eficiency) pathogenesis and provide experimental evidence on CHD (heart qi eficiency)with herbs. Methods:The rats were divided into blank group,model group,sham-operation group,Yiqi Huoxue decoction high、medium、low dose group and western medicine group. CHD (heart qi eficiency)rats model was made by the method of left coronary artery ligation and hyperensort. After 45 d,cardiac muscle tissues were removed and cardiac cell morphological change was observed by HE. Results:Compared with the blank group,model group showed cardiac cell disorder,necrocytosis,too much inflammatory cell infiltration,karyopyknosis deformed or even drop out,increasing the vascular lumen,tissue space widened and filled with a lot of fibrous tissue,horizontal fuzzy fault lines. Compared with model group,apoptosis of Yiqi Huoxue high dose group was significant improved,cardiac cells arranged more neatly,inflammatory cell infiltration and vasodilation was no significant and the degree of fibrosis was reduced. Conclusion:Yiqi Huoxue decoction can restrain and reverse the apoptosis of myocardial cell and alleviate the cellular damage resulted in myocardial ischemia and anoxia.
