建立高效液相法同时测定昙花中含槲皮素、山奈酚和异鼠李素的量.运用Diamonsil C18色谱柱为分析柱,以甲醇-0.4%磷酸水溶液(55∶45)为流动相,流速为1.0 mL/min,检测波长为360 nm.测得槲皮素在5.5~88μg/mL范围内呈良好线性关系,r=0.9999(n=6);山奈酚在4~64μg/mL范围内呈良好线性关系,r=0.9999(n=6);异鼠李素在6.5~104μg/mL范围内呈良好线性关系,r=0.9999(n=6),三者的回收率分别为(99.84±0.23)%(RSD=0.24%)、(100.20±0.52)%(RSD=0.38%)、(99.99±0.12)%(RSD=0.11%).槲皮素、异鼠李素和山奈酚的量分别为0.05、0.026、0.039 mg/g.本方法可同时测定昙花中槲皮素、异鼠李素和山奈酚,操作简单、灵敏、重复性好,适用于昙花及其制剂的质量控制.

A rapid and sensitive HPLC method was developed to detect the content of querce-tin, kaempferol and isorhamnetin in Epiphyllum oxypetalum .Diamond -C18 column was used as chromatographic column , methanol-0.4%phosphoric acid (55∶45) water solution as mobile phase , and the detection wavelength was set at 360 nm, with a flow rate at 1.0 mL/min.There was a good linear relationship when quercetin was in the range of 5.5~88μg/mL, r=0.999 9(n=6), kaempferol was in the range of 4~64 μg/mL, r=0.999 9(n=6) and isorhamnetin was in the range of 6.5~104 μg/mL, r=0.999 9(n=6).The av-erage recoveries were (99.84 ±0.23)% ( RSD=0.24%), (100.20 ±0.52)% ( RSD=0.38%), 99.99 ±0.12% (RSD=0.11%) respectively.The result showed that the con-tent of quercetin, kaempferol and isorhamnetin in Epiphyllum oxypetalum were 0.05, 0. 026, 0.039 mg/g, respectively.The established method was simple , sensitive and repro-ducible to detect the content of quercetin , kaempferol and isorhamnetin in Epiphyllum oxyp-etalum at the same time , which could be applied in the quality control of Epiphyllum oxyp-etalum and its preparations .
