采用高效液相色谱-四极杆-飞行时间质谱法(HPLC-Q-TOF-MS)定性分析了广金钱草中的黄酮类物质,研究了β-乳球蛋白与广金钱草中槲皮素的相互作用,通过直接计算法得到了不同浓度比的β-乳球蛋白与槲皮素的结合数目和结合常数。结果表明,槲皮素能与β-乳球蛋白形成稳定的复合物;单个β-乳球蛋白结合槲皮素的数目随着浓度比的增大而增加,最多为6个,结合常数数量级为104;在高浓度比时,槲皮素与β-乳球蛋白的结合能力比乳糖强,抑制了β-乳球蛋白与乳糖的美拉德反应,有利于提高蛋白质的活性。

High performance liquid chromatography-quadrupole-time of flight mass spectrometry(HPLC-Q-TOF-MS)was established to qualitatively analyze the flavonoid components of Desmodium styracifolium (Osb.)Merr..And the interaction between quercetin andβ-lactoglobulin was analyzed.The binding number and binding constants of quercetin withβ-lactoglobulin at different concentration ratios were measured by direct calculation method.The results indicated that quercetin could form stable complexes withβ-lactoglobulin.The binding ability enhanced with the increasing of concentration ratios ofβ-lactoglobulin and quercetin,and the maximum binding number of quercetin molecules was six when bound toβ-lactoglobulin.The binding constants were measured arround 104 .At high concentration ratios,quercetin possessed better binding ability than lactose when binding toβ-lactoglobulin,and Maillard reaction ofβ-lactoglobulin was inhibited,which may help to im-prove the activity of protein.
