采用对峙培养法从来自土壤的136株细菌中筛选得到对火龙果溃疡病菌有拮抗作用的菌株HTN-5,抑菌带宽9.0 mm。通过形态学、生理生化特性和16s rDNA序列分析,将拮抗菌HTN-5初步鉴定为皮氏类芽孢杆菌Paenibacillus peoriae。从8种培养基中筛选出IFFI培养基最有利于菌株HTN-5形成抑菌活性物质,正交试验优化其成分,得最优配方为:牛肉膏2.50 g/L,蛋白胨10.00 g/L,葡萄糖12.50 g/L,乳糖10.00 g/L,酵母膏2.50 g/L,氯化钠5.00g/L, pH 6.8。

The isolate HTN-5, which antagonized against Neoscytalidium dimidiatum causing pitaya canker, was screened from one hundred and thirty-six bacteria which isolated from the diseased orchard soil. The width of inhibiting zone between HTN-5 and Neoscytalidium dimidiatum reached 9.0 mm. HTN-5 was identified as Paenibacillus peoriae based on morphology, physio-biochemical reactions and 16s rDNA sequences. The IFFI medium was the most suitable medium for isolate HTN-5 to produce antimicrobial active substances, and the optimal medium ingredients, was obtained by the L16 (45) Latin orthogonal experiments, which were comprised of beef extract 2.50 g/L, peptone 10.00 g/L glucose 12.50 g/L, lactose 10.00 g/L, yeast extract 2.50 g/L, sodium chloride 5.00 g/L, pH6.8.
