利用ISSR分子标记,对三叶青全国主分布区24份种质资源的遗传多样性与亲缘关系进行了研究.结果显示:(1)从101个引物里筛选出22个能扩增出清晰具多态性的引物,共扩增出条带169条,片段大小在450~2 200 bp,其中多态性条带77条,多态性比例为46.7%.(2)NTSYSpc2.10e软件分析显示,供试三叶青种质资源遗传多样性较一般,相似系数在0.77~0.99,浙江庆元种质与广西钟山种质亲缘关系最远.(3) UPGMA法聚类分析显示,24份样品被分为4大类,浙江种质全部聚在第Ⅰ类,第Ⅱ类以江西、广西、湖北、湖南等种质为主,湖南怀化与广西钟山为第Ⅲ类,贵州种质单独聚为第Ⅳ类.研究表明,ISSR分子标记适用于三叶青的遗传多样性和亲缘关系分析,研究成果首次从分子水平为合理引种、驯化、保护和利用三叶青野生资源提供了重要的参考依据和数据支持.

Genetic diversity and relationship of 24 germplasms from Tetrastigma hemsleyanum were ana-lyzed by ISSR.The results were as follows:(1) 22 primers which could amplify distinct and polymorphic bands were selected from 101 ones,169 bands (77 polymorphic ones) were amplified whose sizes ranged from 450-2 200 bp,the ratio of polymorphic bands was 46.7%.(2) There was a certain genetic diversity among 24 germ-plasms from Tetrastigma hemsleyanum analyzed by NTsys2.10e,the range of similarity coefficient was from 0.77 to 0.99.Besides,there was the farthest genetic relationship between NO.2 and NO.18 germplasms.(3) 24 sam-ples were divided into 4 groups by UPGMA:all Zhejiang samples were clustered in group Ⅰ,group Ⅱ was based on Jiangxi,Guangxi,Hubei and Hunan germplasms,NO.4 and NO.18 were in groupⅢ,and the Guizhougermplasm was in the Ⅳ group individually.The results indicated that ISSR could be used to analyze genetic diversity and relationship in Tetrastigma hemsleyanum.The conclusion firstly offered valuable references and data at the molecular level for introduction,acclimation,protection and utilization of Tetrastigma hemsleyanum wild resources.
