为了获得适用于发酵生产Nisin的海藻酸钠(A )‐壳聚糖(C )‐海藻酸钠(A )乳链球菌6032的液芯微囊,以其发酵液吸光度值OD600、Nisin的效价为综合指标,探讨了影响ACA乳链球菌6032液芯微囊发酵性能的制备工艺条件.在单因素实验的基础上采用Box‐Behnken模型响应面设计,建立了乳酸菌ACA液芯微囊制备的三元二次回归模型,并确定了其制备因素的最佳水平值为壳聚糖溶解时间2 h、搅拌速度236 r/m in、制备温度33.7℃、滴加速度4.9 mL/min .将此条件下制得的乳酸菌ACA液芯微囊发酵生产Nisin ,吸光度值OD600为0.181, Nisin效价为5242 IU/mL ,该结果与模型预测值拟合良好,说明响应面分析法优化该实验是有效的.
為瞭穫得適用于髮酵生產Nisin的海藻痠鈉(A )‐殼聚糖(C )‐海藻痠鈉(A )乳鏈毬菌6032的液芯微囊,以其髮酵液吸光度值OD600、Nisin的效價為綜閤指標,探討瞭影響ACA乳鏈毬菌6032液芯微囊髮酵性能的製備工藝條件.在單因素實驗的基礎上採用Box‐Behnken模型響應麵設計,建立瞭乳痠菌ACA液芯微囊製備的三元二次迴歸模型,併確定瞭其製備因素的最佳水平值為殼聚糖溶解時間2 h、攪拌速度236 r/m in、製備溫度33.7℃、滴加速度4.9 mL/min .將此條件下製得的乳痠菌ACA液芯微囊髮酵生產Nisin ,吸光度值OD600為0.181, Nisin效價為5242 IU/mL ,該結果與模型預測值擬閤良好,說明響應麵分析法優化該實驗是有效的.

In order to obtain the ACA liquid core microcapsules of lactococcus lactis 6032 for Nisin fermentation ,absorbance value of OD600 and titer unit of Nisin in the broth as compre‐hensive indicators ,the liquid core microcapsules preparation conditions affect on fermentation of L actobacillus was discussed .Based on one‐factor experimental design ,the quadratic model was founded by the Box‐Behnken response surface model .The optimal value of each prepara‐tion condition were chitosan dissolution time 2 h ,stirring speed 236 r/min ,temperature 33 .7℃ ,dropping rate 4 .9 mL/min .Under these conditions ,the absorbance value of OD600 was 0 .181 and the titer unit of Nisin was 5 242 IU/mL ,which is consistent with the predicted value .These showed that it is effective for optimizing preparation conditions by response sur‐face methodology .
