建立了高效液相色谱法同时测定牛奶中十种磺胺类药物残留量。样品用5%高氯酸溶液进行沉淀蛋白,经HC-C18柱(250 mm ×4.6 mm,5μm)分离,甲醇-2%乙酸流动相体系梯度洗脱,用紫外检测器进行检测,检测波长为270 nm,外标法定量。结果表明:10种化合物在0.10~2.0μg/mL浓度范围内线性关系良好,相关系数为0.9921~0.9999。添加水平为0.20,0.40,1.0μg/mL时,平均回收率为81.6%~105%。方法预处理简单,适用于牛奶中磺胺类抗生素药物快速筛查。

A novel method for determination of sulfonamides residues in milk using high performance liquid chromatographic method was established. The samples were simply pretreated by protein precipitation using 5% perchloric acid, and then analyzed on a HC-C18 column. The mobile phase consisted of methanol-2% glacial acetic acid, at a flow-rate of 1. 0 mL/min. The sulfonamide residues were measured by HPLC-UV at 270 nm. In the range of 0. 10~2. 0μg/mL, the target analytes showed good linearity with correlation coefficient ( r ) ranging from 0. 9921 to 0. 9999. At the spike levels of 0. 20, 0. 40, 1. 0 μg/mL, satisfactory recoveries ranged from 81. 6% to 105%. The method is simple, rapid, robust and is proved to be suitable for detecting the sulfonamide residues in milk.
