采用苯酚-浓硫酸紫外分光光度法测定多糖含量。通过单因素试验研究料液比、提取水温和提取时间对茯苓超声提取效果的影响,利用正交试验以干浸膏得率、多糖得率为评价指标,优化提取工艺条件。最后确定最佳提取工艺为A2 B3 C2,即茯苓粉末以料液比1:40在水温80℃下超声提取30 min。经验证结果可靠,超声提取茯苓工艺简便安全,稳定可行,多糖得率可达2.87%,干浸膏得率4.01%。

Phenol-sulfuric acid spectrophotometric method was used to determine the content of polysaccharide. Solid-liquid ratio, temperature and extraction time on the content of poria cocos polysaccharide in extracts were investigated by one-factor-at-a-time method, and with dry extract yield and polysaccharide content as evaluation indicator, the orthogonal test was used to optimize the extraction process conditions. Finally, the optimum extraction process was A2 B3 C2 , which meant the solid-liquid ratio was 1:40, the temperature was 80 ℃ and the ultrasonic extraction time was 30 min. The process was a proven and reliable result, which was simple and safe, stable and feasible with the dry extract yield of 2. 87% and the polysaccharide content of 4. 01%.
