为降低烟叶中有害成分,提高其燃吸品质,从片烟样品中筛选得到一株果胶酶高产菌株克雷白氏杆菌(Klebsiella sp.),并对此菌株的发酵培养基进行优化.首先应用单因素实验考察碳源种类及浓度、氮源种类及浓度和发酵初始 pH.在单因素实验的基础上,进行响应面分析优化,确定了最佳发酵条件为:烟末添加量3.30%,硫酸铵添加0.35%,初始 pH7.2.经摇瓶发酵验证,当初始 pH7.2,烟末添加量3.30%,硫酸铵添加0.35%时,该菌株发酵产果胶酶活力最高,达到15.07 U/ml.

In order to reduce the harmful components in tobacco and improve its smoking quality,a pecti-nase produced strain was isolated from tobacco,and identified as Klebsiella sp.Different conditions inclu-ding initial pH,carbon source,nitrogen source for the enzyme production were studied.Base on the results of single factor experiment,the response surface method was applied to determine the combined condition. The result showed that:tobacco powder quantity 3.30%,ammonium sulfate 0.35%,initial pH7.2 was the optical condition;the fermentation production of pectinase activity reached the highest,14.98 U/ml.This result was verified by flask fermentation experiment with optimized condition.Pectinase activity reached as high as 15.07 U/ml.
