目的::研究创伤后应激障碍(PTSD)大鼠肠系膜和软脑膜微循环的改变及其机制。方法:成年健康雄性 SD 大鼠18只,随机分为正常对照组(n=6)和 PTSD 组(n=12)。采用单程延长刺激法复制 PTSD 大鼠模型,通过旷场实验检测大鼠的行为学改变;采用活体微循环观测技术观察大鼠肠系膜及软脑膜微血管内径和血流速度的改变;采用 ELISA 检测大鼠血浆去甲肾上腺素(NA)、内皮型一氧化氮合酶(eNOS)和内皮素-1(ET-1)水平。结果:与正常对照组比较,PTSD 组大鼠在旷场中穿行格数及直立次数显著减少(P <0.01);肠系膜及软脑膜细动脉、细静脉管径明显变细(P <0.05),血流速度趋缓;血浆 NA 和 ET-1含量显著升高(P <0.01),但 eNOS 水平无显著变化(P >0.05)。结论:PTSD 大鼠存在肠系膜和软脑膜微循环障碍。

목적::연구창상후응격장애(PTSD)대서장계막화연뇌막미순배적개변급기궤제。방법:성년건강웅성 SD 대서18지,수궤분위정상대조조(n=6)화 PTSD 조(n=12)。채용단정연장자격법복제 PTSD 대서모형,통과광장실험검측대서적행위학개변;채용활체미순배관측기술관찰대서장계막급연뇌막미혈관내경화혈류속도적개변;채용 ELISA 검측대서혈장거갑신상선소(NA)、내피형일양화담합매(eNOS)화내피소-1(ET-1)수평。결과:여정상대조조비교,PTSD 조대서재광장중천행격수급직립차수현저감소(P <0.01);장계막급연뇌막세동맥、세정맥관경명현변세(P <0.05),혈류속도추완;혈장 NA 화 ET-1함량현저승고(P <0.01),단 eNOS 수평무현저변화(P >0.05)。결론:PTSD 대서존재장계막화연뇌막미순배장애。
Objective:To investigate the effect of post-traumatic stress disorder (PTSD)on microcirculation of mesentery and cerebral pia mater in rats and the underlying mechanisms.Method:Sprague-Dawley (SD)rats were randomly divided into two groups:control group (n=6)and PTSD group (n=12).The PTSD model was rep-licated by using the single prolonged stress method.On the 14th day post-stress,the changes of animals'behavior was evaluated via open-field;the microcirculation of mesentery and cerebral pia mater of rats was observed using the microscope;the plasma noradrenaline (NA),endothelial nitric oxide synthase (eNOS)and endothelin-1 (ET-1 ) were measured by using enzyme linked immunosorbent assay (ELISA).Results:Compared with the control rats,the numbers of crossing and rearing of the PTSD rats were decreased significantly in the open-field (P <0.01);the di-ameters of arteriole and veinule in the mesentery and cerebral pia mater from the PTSD rats were significantly thin-ner (P <0.05),the blood flow velocity showed the trend of slowed down;the plasma levels of NA and ET-1 were elevated significantly (P <0.01);there was no significant difference in the plasma level of eNOS (P >0.05).Con-clusion:PTSD induces microcirculation dysfunction in mesentery and cerebral pia mater in rats.
