目的:检测B7家族在非霍奇金淋巴瘤( NHL)中的表达,阐明其在NHL中的作用及意义。方法应用密度梯度离心法分离健康志愿者外周血单个核细胞( PBMCs),实时荧光定量PCR( RQ-PCR)检测B7家族PD-L1和PD-L2 mRNA在B细胞淋巴瘤(B-NHL)、T细胞淋巴瘤(T-NHL)及NK/T细胞淋巴瘤(NKTL)细胞系中的表达水平。结果 PD-L1和PD-L2 mRNA在NHL各细胞系中呈不同程度表达,T-NHL及NKTL细胞系中其表达量均明显高于正常PBMCs(P<0.05),而B-NHL细胞系中PD-L1和PD-L2 mRNA表达量与正常PBMCs相比无明显差异( P>0.05)。结论在NHL中, B7家族中PD-L1及PD-L2可能参与了NKTL及T-NHL肿瘤的免疫逃逸,有望成为NKTL及T-NHL肿瘤免疫治疗的新靶点。

ObjectiveTo detect the expression of B7 family molecules and illuminatetheir significance innon-Hodgkin’s lymphoma (NHL).Methods Peripheralbloodmonocytecells (PBMCs) were separated by density gradient centrifugation from peripheral blood samples of healthy volunteers. Real-time fluorescence quantitative PCR (RQ-PCR) was used to determine the mRNA expression levels of PD-L1 and PD-L2 in the cell lines of B cell lymphoma, T cell lymphoma and NK/T cell lymphoma (NKTL).Results PD-L1 and PD-L2 were expressed in NHL cell lines at different mRNA levels. Their expression was significantly higher in T cell lymphoma and NKTL cell lines thaninnormal PBMCs (P<0.05). However, there was no significant difference in theirexpression between B cell lymphoma cell lines and PBMCs (P>0.05).Conclusion B7 family molecules such as PD-L1 and PD-L2 may be involved in the tumor immune escape in NKTL and T cell lymphoma, and they might be novel targets for NHL immunotherapy.
