基于1H NMR的代谢组学方法对灌胃给药赭石的成年Wistar大鼠血清进行分析,运用主成分分析模式识别方法对所得数据进行处理,并对给药大鼠血清进行生化指标检测.研究结果表明,大鼠体内?-羟异丁酸、乙酸、丙酮、胆碱、甘油磷脂酰胆碱、葡萄糖、乳酸、低密度脂蛋白、极低密度脂蛋白和脂质等内源性代谢物浓度发生明显变化,可作为赭石的特征代谢物.2 g/kg和5 g/kg体重剂量赭石使大鼠机体产生大量活性氧化物(ROS),造成过氧化损伤,导致能量代谢和糖代谢紊乱,糖酵解反应增强,并且对肝功能造成了影响.

Wistar rats were intragastrically administered with different doses (0, 2, 5 g/kg body weight) of haematitum.1H NMR-based metabonomic techniques were used to analyze the metabolic profiles of the serum samples collected from the treated rats. Significant treatment-related changes were observed for the levels of?-hydroxybutyate, acetate, acetone, choline, glycerophosphorylcholine, glucose, lactate, low density lipoprotein, very low density lipoprotein and lipids. Such biochemical changes indicated that haematitum treatment at the dose of 2 or 5 g/kg body weight gave rise to reactive oxygen species (ROS) and caused peroxidation damage. It is concluded that haematitum treatment affects glucose, lipid and energy metabolism in rats.
