目的:观察四神丸对脾肾阳虚型溃疡性结肠炎大鼠中血清转化生长因子β1( TGFβ1)、白介素6(IL-6)及结肠组织中Toll样受体4(TLR-4) mRNA含量的变化,探讨四神丸对脾肾阳虚型溃疡性结肠炎的作用机制。方法:采用番泻叶灌胃+氢化可的松注射液腹腔注射+2、4、6-三硝基苯磺酸( TN-BS)/乙醇法灌肠建立脾肾阳虚型溃疡性结肠炎大鼠模型。将60只wistar大鼠随机分为空白组、模型组、四神丸组及柳氮磺嘧啶( SASP )组,经药物干预后分别采用酶联免疫法检测各组大鼠血清中的TGFβ1、IL-6含量及半定量反转录-聚合酶链反应检测各组大鼠结肠组织中的TLR-4 mRNA含量。结果:与空白组比较,模型组大鼠中TGFβ1含量降低(P<0.05),IL-6含量升高(P<0.05);与模型组比较,各治疗组大鼠中TGFβ1含量升高(P<0.05),IL-6含量降低(P<0.05)。与空白组比较,模型组大鼠结肠组织TLR-4 mRNA表达显著增强(P<0.05);各治疗组TLR-4 mRNA表达减弱(P<0.05)。结论:四神丸能够通过下调IL-6和升高TGFβ1的表达,降低结肠组织中TLR-4 mRNA的含量,抑制炎症反应,调节肠上皮免疫系统,恢复肠粘膜微环境的免疫稳态,达到治疗UC的目的。
目的:觀察四神汍對脾腎暘虛型潰瘍性結腸炎大鼠中血清轉化生長因子β1( TGFβ1)、白介素6(IL-6)及結腸組織中Toll樣受體4(TLR-4) mRNA含量的變化,探討四神汍對脾腎暘虛型潰瘍性結腸炎的作用機製。方法:採用番瀉葉灌胃+氫化可的鬆註射液腹腔註射+2、4、6-三硝基苯磺痠( TN-BS)/乙醇法灌腸建立脾腎暘虛型潰瘍性結腸炎大鼠模型。將60隻wistar大鼠隨機分為空白組、模型組、四神汍組及柳氮磺嘧啶( SASP )組,經藥物榦預後分彆採用酶聯免疫法檢測各組大鼠血清中的TGFβ1、IL-6含量及半定量反轉錄-聚閤酶鏈反應檢測各組大鼠結腸組織中的TLR-4 mRNA含量。結果:與空白組比較,模型組大鼠中TGFβ1含量降低(P<0.05),IL-6含量升高(P<0.05);與模型組比較,各治療組大鼠中TGFβ1含量升高(P<0.05),IL-6含量降低(P<0.05)。與空白組比較,模型組大鼠結腸組織TLR-4 mRNA錶達顯著增彊(P<0.05);各治療組TLR-4 mRNA錶達減弱(P<0.05)。結論:四神汍能夠通過下調IL-6和升高TGFβ1的錶達,降低結腸組織中TLR-4 mRNA的含量,抑製炎癥反應,調節腸上皮免疫繫統,恢複腸粘膜微環境的免疫穩態,達到治療UC的目的。
목적:관찰사신환대비신양허형궤양성결장염대서중혈청전화생장인자β1( TGFβ1)、백개소6(IL-6)급결장조직중Toll양수체4(TLR-4) mRNA함량적변화,탐토사신환대비신양허형궤양성결장염적작용궤제。방법:채용번사협관위+경화가적송주사액복강주사+2、4、6-삼초기분광산( TN-BS)/을순법관장건립비신양허형궤양성결장염대서모형。장60지wistar대서수궤분위공백조、모형조、사신환조급류담광밀정( SASP )조,경약물간예후분별채용매련면역법검측각조대서혈청중적TGFβ1、IL-6함량급반정량반전록-취합매련반응검측각조대서결장조직중적TLR-4 mRNA함량。결과:여공백조비교,모형조대서중TGFβ1함량강저(P<0.05),IL-6함량승고(P<0.05);여모형조비교,각치료조대서중TGFβ1함량승고(P<0.05),IL-6함량강저(P<0.05)。여공백조비교,모형조대서결장조직TLR-4 mRNA표체현저증강(P<0.05);각치료조TLR-4 mRNA표체감약(P<0.05)。결론:사신환능구통과하조IL-6화승고TGFβ1적표체,강저결장조직중TLR-4 mRNA적함량,억제염증반응,조절장상피면역계통,회복장점막미배경적면역은태,체도치료UC적목적。
Objective:To observe the influence of Sishen pill on serum of transforming growth factor beta 1 (TGFβ1), in-terleukin 6 (IL-6) and colonic tissue of toll -like receptor 4 (TLR-4) mRNA content changes and identify its under-lying mechanism of action .Methods:Use lavage of senna +intraperitoneal injection of hydrocortisone injection +enema of 2, 4, 6-trinitrobenzene sulfonic acid (TNBS)/ethanol to establish the rat models of ulcerative colitis with spleen -kidney deficiency type .60 wistar rats were randomly divided into a blank group ,a model group , a Sishen pill group and a salazosulfadimidine (SASP) group.After drug intervention, serums of TGF β1 and IL-6 were detected by ELISA, and the colonic tissue of TNF -αmRNA was detected by Sq RT -PCR.Results: Compared with those in the blank group,in the model group rats’ TGFβ1 was decreased (P<0.05),but IL-6 content was increased (P<0.05);com-pared with those in the model group ,in the treatment group TGF β1 content was increased (P<0.05),but IL-6 was decreased (P<0.05).Compared with that in the blank group , the expression of the model group ’s TLR-4 mRNA of colonic tissue was significantly enhanced (P<0.05);the expression of each treatment group ’s TLR-4 mRNA was de-creased (P<0.05).Conclusion:Sishen pill can decrease the expression of IL -6, increase the expression of TGF β1, reduce content of TLR-4 mRNA in colonic tissue ,and then it can inhibit the reaction of inflammation , regulate intesti-nal epithelial immune system , restore intestinal mucosal immune steady -state microenvironment , then achieve the pur-pose of the treatment of UC .
