目的:观察生地含药血清对高糖刺激下人肾小球系膜细胞(HRMC)的作用。方法制备生地含药血清,通过 LC-MS检测技术,鉴别入血成分;MTT 法检测 HRMC 增值抑制率;试剂盒检测细胞上清中 SOD、MDA 的含量变化;ELISA 法检测HRMC 中纤维连接蛋白(FN)、Ⅳ型胶原(ColⅣ);流式法检测细胞周期分布;免疫荧光检测 HRMC 中 TGF-β1的表达。结果生地含药血清中含有水苏糖、梓醇,含药血清中效应成分可抑制高糖诱导的 HRMC 增殖,减少细胞外基质(ECM)的增生,下调 TGF-β1的 mRNA 表达。结论生地可能通过调节 TGF-β1的表达来抑制 ECM 的分泌从而改善糖尿病肾病的发生发展。

목적:관찰생지함약혈청대고당자격하인신소구계막세포(HRMC)적작용。방법제비생지함약혈청,통과 LC-MS검측기술,감별입혈성분;MTT 법검측 HRMC 증치억제솔;시제합검측세포상청중 SOD、MDA 적함량변화;ELISA 법검측HRMC 중섬유련접단백(FN)、Ⅳ형효원(ColⅣ);류식법검측세포주기분포;면역형광검측 HRMC 중 TGF-β1적표체。결과생지함약혈청중함유수소당、재순,함약혈청중효응성분가억제고당유도적 HRMC 증식,감소세포외기질(ECM)적증생,하조 TGF-β1적 mRNA 표체。결론생지가능통과조절 TGF-β1적표체래억제 ECM 적분비종이개선당뇨병신병적발생발전。
OBJECTIVE To investigate the effects of Rehmanniae-medicated serum on human glomerular mesangial cells (HRMC)stimulated with high concentration of glucose.METHODS Rehmanniae-medicated serum was prepared and analyzed for its serum components using LC-MS technology.MTT assay was used to detect the inhibitory rate of HRMC.Cell superna-tant levels of SOD and MDA were measured by kits.ELISA assays were used to determine the contents of fibronectin and type IV collagen.Flow cytometry was used to analyze cell cycle distributions.Immunofluorescence was used to detect the expres-sion of TGF-β1 in HRMC.RESULTS Rehmanniae-medicated serum contains catalpa alcohol and stachyose.These active com-ponents could inhibit high level glucose-induced HRMC proliferation,reduced the production of extracellular matrix,and downregulated the mRNA expression of TGF-β1 .CONCLUSION Rehmanniae could inhibit ECM production by regulating TGF-β1 so as to prevent the development of diabetic nephropathy.
