Fushen Decotion%diabetic nephropathy%db/db mice
目的:观察复肾汤对 db/db 小鼠肾脏病变的影响。方法正常对照组为雄性 C57小鼠8只,常规饲养。雄性 db/db 鼠24只,正常饲养1周,血糖稳定升高4周后分为模型组、复肾汤小剂量组(L1)、复肾汤大剂量组(L2)。模型组灌胃给予生理盐水;L1组按 FST 13 mg/(kg·d)灌胃;L2组按 FST 39 mg/(kg·d)灌胃;连续8周,定期测体质量、血糖。实验结束后,采集血清,检测 T-chol、BUN、SOD、MDA 等生化指标;轻取肾脏,免疫组织化学法检测肾组织 VEGF、FN 的表达,光镜下观察肾脏组织病理变化。结果与模型组比较,L1、L2组的 T-chol、BUN、MDA 明显降低(P <0.05~0.01),SOD 明显升高(P <0.05), VEGF、FN 表达明显下调,病理学评分明显降低(P <0.05)。结论复肾汤可以明显改善 db/db 小鼠肾脏损害,具有延缓糖尿病肾病病情发展的作用。

목적:관찰복신탕대 db/db 소서신장병변적영향。방법정상대조조위웅성 C57소서8지,상규사양。웅성 db/db 서24지,정상사양1주,혈당은정승고4주후분위모형조、복신탕소제량조(L1)、복신탕대제량조(L2)。모형조관위급여생리염수;L1조안 FST 13 mg/(kg·d)관위;L2조안 FST 39 mg/(kg·d)관위;련속8주,정기측체질량、혈당。실험결속후,채집혈청,검측 T-chol、BUN、SOD、MDA 등생화지표;경취신장,면역조직화학법검측신조직 VEGF、FN 적표체,광경하관찰신장조직병리변화。결과여모형조비교,L1、L2조적 T-chol、BUN、MDA 명현강저(P <0.05~0.01),SOD 명현승고(P <0.05), VEGF、FN 표체명현하조,병이학평분명현강저(P <0.05)。결론복신탕가이명현개선 db/db 소서신장손해,구유연완당뇨병신병병정발전적작용。
OBJECTIVE To observe the effects of Fushen Decotion on renal lesion in db/db mice.METHODS 8 male C57 mice were chosen as normal control group,conventional breeding.24 male db/db mice with high blood glucose level were di-vided into model group,FST(L1) group and FST(L2) group.Model group mice were given normal saline;L1 and L2 mice were treated with FST 13 g/kg and 39 g/kg,respectively.The weight and blood sugar of mice were measured regularly for 8 weeks.After the experiment,the serum was collected and biochemical parameters,such as T-chol,BUN,SOD and MDA, were detected.Then the kidneys were taken,the expression of VEGF and FN in renal tissues were detected by immunohisto-chemical method,and the renal lesions were observed under light microscope.RESULTS Compared with the model group,T-chol,BUN and MDA of L1 and L2 groups were significantly decreased(P <0.05~0.01),SOD was increased significantly (P <0.05),expression of VEGF and FN decreased significantly,and pathology score decreased significantly (P < 0.05). CONCLUSION Fushen Decotion can significantly improve the db/db mice renal damage and delay the development of diabetic nephropathy.
