目的:研究生脉、地塞米松联合局部低温对大鼠睾丸扭转复位后生精细胞凋亡的影响。方法将48只青春期标准SD大鼠随机分为6组( A、B、C、D、E、F组),每组8只。6组大鼠分别扭转左侧睾丸720°持续2 h,建立单侧睾丸扭转模型。 A组:常温+生理盐水;B组:常温+生脉+地塞米松;C组:低温+生理盐水;D组:低温+地塞米松;E组:低温+生脉;F组:低温+生脉+地塞米松。术后48 h采集睾丸,通过HE染色光镜观察睾丸组织病理学改变,用原位缺口末端标记法( TUNEL)法检测睾丸生精细胞的凋亡。结果 B、C、D、E组,睾丸组织可见部分生精细胞脱落;F组,小鼠睾丸内出现生精细胞脱落现象。凋亡指数(AI),A组(35.13±2.21)明显高于B组(18.12±5.23)、C 组(17.13±3.03)、D组(17.05±1.03)、E 组(16.39±2.02;均 P <0.05)及 F 组(9.06±1.53;P <0.01)。结论睾丸扭转复位后缺血再灌注损伤可导致生精细胞凋亡增加;局部低温联合生脉、地塞米松对睾丸扭转复位后生精细胞凋亡具有良好保护作用。

목적:연구생맥、지새미송연합국부저온대대서고환뉴전복위후생정세포조망적영향。방법장48지청춘기표준SD대서수궤분위6조( A、B、C、D、E、F조),매조8지。6조대서분별뉴전좌측고환720°지속2 h,건립단측고환뉴전모형。 A조:상온+생리염수;B조:상온+생맥+지새미송;C조:저온+생리염수;D조:저온+지새미송;E조:저온+생맥;F조:저온+생맥+지새미송。술후48 h채집고환,통과HE염색광경관찰고환조직병이학개변,용원위결구말단표기법( TUNEL)법검측고환생정세포적조망。결과 B、C、D、E조,고환조직가견부분생정세포탈락;F조,소서고환내출현생정세포탈락현상。조망지수(AI),A조(35.13±2.21)명현고우B조(18.12±5.23)、C 조(17.13±3.03)、D조(17.05±1.03)、E 조(16.39±2.02;균 P <0.05)급 F 조(9.06±1.53;P <0.01)。결론고환뉴전복위후결혈재관주손상가도치생정세포조망증가;국부저온연합생맥、지새미송대고환뉴전복위후생정세포조망구유량호보호작용。
Objective To investigate the protective effect of hypothermia union Shengmai and dexamethasone on spermatogenic cell apoptosis of torsioned testes in rats.Methods Forty-eight adolescence male Spra-gue-Dawley rats were random divided into 6 groups.Each group of 8 rats, six group rats were left testicular torsion 720°for 2 h, establish a u-nilateral testicular torsion model.Group A:normal temperature+saline;group B:normal+Shengmai+dexamethasone; group C:low temperature+saline;group D:low temperature+dexamethasone; group E:low tem-perature+Shengmai;group F:Shengmai+dexamethasone+low tempera-ture.The testicles were collected after 48 h, the testicular histopathologi-cal changes were observed by HE staining light microscopy.The apopto-tic spermatogenic cells law were detected by TdT-mediated dUTP nick-end labeling ( TUNEL) .Results Group B, C, D and E testicular tissue pathology damage visible parts of pure cell falls off born.The born small tube of testicular sperm cells in group F is born slightly off, general structure and tidy.Apoptosis index ( AI ) in group A ( 35.13 ±2.21 ) was higher than that group B ( 18.12 ±5.23 ) , group C ( 17.13 ±3.03 ) , group D ( 17.05 ±1.03 ) , group E ( 16.39 ±2.02;all P<0.05 ) and group F ( 9.06 ±1.53;P <0.01 ) . Conclusion Testicular torsion reduction ischemia-reperfusion injury can lead to life essence cell apoptosis increase.Hypothermia union Shengmai and dexa-methasone on spermatogenic cell apoptosis of torsioned testes in rats have good protection.
