目的 应用酶渗漉法和醇渗漉法分别提取美人草(颠茄草),考察两种溶剂提取对其中主要活性物质莨菪碱的提取率,优选最佳提取方法 . 方法 用pH值为2.0、体外消化活力为24~30μ· mL -1的胃蛋白酶溶液为溶剂渗漉美人草,以80%乙醇渗漉液做对照,用高效液相色谱法测定两组提取液中莨菪碱的含量,比较其提取率. 结果 酶渗漉提取法莨菪碱提取率为4.79mg· g-1,醇渗漉提取法提取率为3.52mg· g -1 ,酶渗漉法比醇渗漉法提取率高36.1%. 结论 酶渗漉法提取美人草能显著提高主要活性物质莨菪碱的提取率,可用于美人草活性物质的提取.

ABJECTIVE Enzyme percolation method and alcohol percolation method dwere utilized to extract beauty herbal( Atropa belladonna L).Extraction rates of the main active substance-hyoscyamine by two methods were investigated.The bestter extraction method was selected.METHODS In the experimental group, the pepsin solu-tion,pH was 2.0,in vitro digestion energy was 24~30μ· mL-1 ,was utilized as solvent to percolate beauty herbal.In the control group,the 80%ethanol solution was utilized to percolate beauty herbal.High performance liquid chroma-tography was utilized to test the content of hyoscyamine in extract of two groups and extraction rates of two groups were compared.RESULTS The extraction rate of hyoscyamine in experimental group was 4.79 mg · g-1 , it was 36.1%higher than that of control group which was 3.52 mg · g-1 .CONCLUSION Enzyme percolation method can significantly increase the extraction rate of the main active substance-hyoscyamine in beauty herbal.This method can be used to extract the active substance in beauty herbal.
